摘要
目的:探讨膀胱癌中长链非编码RNA(lnc RNA)TUC338的表达及作用机制。方法 :收集安徽医科大学一附院收治的30例膀胱癌手术患者的癌变组织和癌旁组织,通过实时定量PCR检测TUC338的表达水平并分析其临床病理特征之间的关联。运用RNA干扰(si-TUC338)的方法检测敲减TUC338,采用CCK8和半胱天冬酶3酶联免疫吸附测定法的方法分别检测TUC338对膀胱癌细胞的增殖、凋亡的影响。结果:相对于膀胱癌旁组织,TUC338 m RNA在膀胱癌组织中高表达。敲减TUC338后,细胞增殖被抑制,细胞凋亡增加。结论:TUC338是一个潜在的治疗膀胱癌的药物作用靶点。
Objective To detect the expression and roles of long non-coding RNA(lnc RNA) transcribed ultra-conserved region 338(TUC338) on bladder cancer Methods Thirty pairs of bladder urothelial carcinomas and adjacent normal specimens were collected.Realtime PCR was used to detect the expression of TUC338 and to analyze the correlation between its expression and clinical pathological features in the specimens.Si-TUC338 was transfected to bladder cancer cells to knockdown TUC338 expression.CCK8 assay and the caspase3 enzyme-linked immunosorbent assay(ELISA) was used to test the proliferation and apoptosis of bladder cancer cells.Results The expression of TUC338 was significantly unregulated in bladder cancer samples compared with adjacent normal samples.Si-TUC338 could significantly inhibit the proliferation and increase apoptosis.Conclusions TUC338 is a new drug candidate for bladder cancer.
