丹参药渣中丹参酮类化学成分的提取富集研究及其利用途径分析 被引量:9

摘要 目的:对丹参药渣中丹参酮类成分的提取富集工艺进行优化,并进一步对其开发利用进行分析。方法:采用L9(3-4)正交试验设计,以总丹参酮的含量为评价指标,优选丹参药渣的最佳提取工艺条件;采用溶剂沉淀法纯化富集丹参酮类成分。结果:丹参药渣中丹参酮类成分的最佳提取工艺为乙醇体积分数为90%,10倍乙醇用量,提取2 h,提取2次;通过水沉淀法可以去除丹参粗提物中大多数的水溶性成分,纯化制备丹参酮类化学成分,其中提取物中总丹参酮的含量为1.752%,是原提取物含量的7.5倍。结论:本文优选了丹参药渣中总丹参酮的最佳提取工艺,并采用水沉淀去除部分水溶性成分,从而富集纯化丹参酮类成分。建立了采用HPLC同时测定丹参药渣中总丹参酮含量的方法,操作简便快速、重现性好,为丹参药渣的资源化利用及质量控制提供科学依据与参考。 Objective: To optimize the extraction and enrichment process of tanshinones in the Salvia miltiorrhiza residues. and analysis the development and utilization of Salvia miltiorrhiza residues for further. Methods: The extraction conditions of Salvia dregs was optimized by using the L9(3-4) orthogonal test,with the amount of total tanshinone as evaluation index. Tanshinones were purified and accumulated by solvent precipitation. Results: The optimum parameters for extraction process of the total tanshinone were as follows: 10 times amount of 90% ethanol,extracting for 2 times,each time 2 h. Water precipitation could enrich salvianolic acids. The content of the total tanshinone was 1. 752%,7. 5 times as in original crude extracts. Conclusion: This paper select the best extracting technology of tanshinones in in the Salvia miltiorrhiza residues,and uses the water precipitation to remove part of the water soluble component,purification and enrichment tanshinones. and established a method for determination of tanshinones from Salvia miltiorrhiza residues in content by HPLC method,and this method is simple,fast and reproducible,and can be a reference for the quality control of the extract of Salvia miltiorrhiza residues. and provide scientific basis and reference for the resource utilization and quality control of Salvia miltiorrhiza residues.
