摘要
目的:建立高效液相色谱法同时测定十味玉泉片中葛根素、甘草苷、甘草酸铵和五味子醇甲4种有效成分的含量。方法:色谱柱为ZORBAX SB-C18柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸水溶液进行梯度洗脱;流速为1.0 m L·min^(-1);柱温为30℃;检测波长(0~8 min在250 nm波长下检测葛根素,8~14 min在276 nm波长下检测甘草苷,14~25 min在250 nm波长下检测甘草酸铵,25~40min在216 nm波长下检测五味子醇甲)。结果:4个有效成分的线性范围分别为葛根素68.05~680.5μg·m L^(-1)(r=0.999 9)、甘草苷22.06~220.6μg·m L~(^(-1))(r=0.999 8)、甘草酸铵24.98~249.8μg·m L^(-1)(r=0.999 4)、五味子醇甲2.184~21.84μg·m L^(-1)(r=0.998 9),平均加样回收率分别为98.83%、97.84%、98.09%、97.20%,RSD分别为0.71%、0.69%、0.80%、0.99%(n=6)。结论:该方法简便、准确、灵敏度高、重复性好,可用于同时测定十味玉泉片中上述4个有效成分的定量测定,控制产品质量。
Objective: To establish an HPLC method for simultaneous determination of four active components( puerarin; liquiritin; ammonium glycyrrhetate; schisandrin) in the Shiwei Yuquan tablets. Methods: Zorbax SB-C18( 250 mm × 4. 6 mm,5 μm) column was used with mobile phase consisting of water containing 0. 1% phosphoric acid and acetonitrile in gradient mode. The flow rate was 1. 0 m L·min^-1,The column temperature was 30 ℃. Puerarin was detected at250 nm,liquiritin was detected at 276 nm,ammonium glycyrrhetate were detected at 250 nm,and schisandrin was detected at216 nm. Results: There was good linear relationship between the concentration of puerarin, liquiritin, ammonium glycyrrhetate,schisandrin,and peak area within the concentration range of 68. 05- 680. 5 μg·m L^-1( r = 0. 999 9),22. 06-220. 6 μg·m L^-1( r = 0. 999 8),24. 98- 249. 8 μg·m L^-1( r = 0. 999 4),2. 184- 21. 84 μg·m L^-1( r = 0. 998 9). The average recovery( RSD) was 98. 83%( 0. 71%),97. 84%( 0. 69%),98. 09%( 0. 80%),97. 20%( 0. 99%),respectively( n = 6). Conclusion: The established method is accurate,highly sensitive and well reproducible,which can be used for the determination of the four active ingredient in the Shiwei Yuquan tablets,control product quality.
