水对离子液体溶解再生蚕蛹蛋白的影响 被引量:1

Effect of Water on Dissolution and Regeneration of Silkworm Pupa Protein by Using Ionic Liquids

摘要 以蚕蛹粉为原料,考察了含水量对离子液体1-烯丙基-3-甲基咪唑氯([Amim]Cl)溶解再生蚕蛹蛋白的影响,并对再生蚕蛹蛋白的结构和热稳定性进行了分析.结果表明,水会破坏[Amim]Cl的氢键结构,降低对蚕蛹粉的溶解再生效率;不含水的[Amim]Cl溶解再生蚕蛹蛋白的效果最好,在溶解温度90℃、时间24 h、乙醇为再生剂的条件下,蚕蛹粉溶解率为86.5%,蚕蛹蛋白再生率达59.6%,再生物为蛋白质,并具有α螺旋、β折叠二级结构,且β折叠结构比例大于α螺旋结构;离子液体含水量对再生蛋白的结构没有明显影响,再生蛋白的热稳定性比蚕蛹粉稍有提高. The effect of water content in 1-allyl-3-methylimidazolium chloride([Amim Cl]) on the dissolution and regeneration of silkworm pupa protein(SPP) was explored with silkworm pupa as the raw materials. The structure and thermostability of regenerated SPP were analyzed. The results suggested that water could break the hydrogen bonding structure of [Amim Cl] and exert a negative effect on the dissolution rate of silkworm pupa powder and the yield of regenerated SPP. Anhydrous [Amim]Cl exhibited the best effect. When the dissolution mechanism was conducted in [Emim]Cl at 90 ℃ for 24 h and ethanol bath was used for the regeneration process, the dissolution rate of silkworm pupa was 86.5% and the yield of regenerated SPP could reach 59.6%. The regenerated material was SPP, the secondary structure of which was constituted of α-helix and β-sheet. However, β-sheet showed a higher intensity than α-helix. The water content of ionic liquid had no obvious effect on the structure of regenerated protein, while the thermostability of SPP was slightly improved than silkworm pupa powders.
