从唾液中提取脱氧核糖核酸扩增血红蛋白β基因

摘要 目的:探索一种对人体无害、无痛、操作简便的获取基因组脱氧核糖核酸(deoxyribonucleic acid,DNA)的方法。方法:从唾液中提取基因组DNA,应用聚合酶链式反应(polymerase chain reaction,PCR)扩增血红蛋白β基因,采用含溴乙锭的琼脂糖凝胶电泳鉴定扩增产物。结果:从唾液中提取基因组DNA能够扩增出血红蛋白β基因。结论:唾液作为获取基因组脱氧核糖核酸的简易来源,取样方法简便、快捷,更适用于疾病研究中大规模群体筛查。 Objective: To explore a harmless, painless and easily - operated method for genomic deoxyribonucleic acid (DNA). Methods: Genomic DNA was extracted from saliva. Polymerase chain reaction (PCR) was applied to amplify hemoglobin 13 gene, and the amplification products were identified by agarose gel electrophoresis containing ethidium bromide. Results:The genomic DNA extracted from saliva can amplify the hemo- globin 13 gene. Conclusion : Saliva is a simple source of genomic DNA. The method is simple and fast, and may be used for screening on a large scale.
