益肾活血解毒汤对冠心病合并糖尿病患者炎性因子蛋白表达作用的研究

Effect of Yishen Huoxue Jiedu Decoction on the Expression of Inflammatory Factors in Patients with Coronary Heart Disease Complicated with Diabetes Mellitus

目的:观察益肾活血解毒汤对血小板活化因子(Platelet-activatingfactor,PAF)诱发的内皮细胞炎性因子蛋白表达的作用。方法:采用随机化区组法将所观察的冠心病合并糖尿病患者分为A组(未常规服用阿托伐他汀20mgqd),B组(常规预口服阿托伐他汀20mgqd>7d),C组(常规予口服阿托伐他汀20mgqd>7d并预加用益肾活血解毒汤150ml bid)各15人。清晨空腹抽取三组患者血液化验肝肾功能、血糖、血脂、凝血功能等相关指标,酶联免疫吸附测定法(ELISA)检测三组血清中单核细胞白细胞介素-6(interleukin-6,IL-6)、白介素-1β(interleukin-1β,IL-1β)和肿瘤坏死因子-α(Tumor Necrosis Factor-a,TNF-a)蛋白表达水平。A组和C组抽血后当天加用益肾活血解毒汤150ml bid,三组连续口服4周后再次检查相同指标。结果:与C组相比较,A、B组中TNF-α及IL-1β、IL-6蛋白表达水平明显高于C组(P<0.05)。结论:益肾活血解毒汤抑制PAF诱导的血清中单核细胞IL-6、IL-1β和TNF-a蛋白表达的升高。益肾活血解毒汤合并阿托伐他汀能够更好的抑制PAF所诱导的内皮细胞炎症介质表达。

Objective： To observe the effect of Yishen Huoxue Jiedu decoction on expression of inflammatory factor protein induced by platelet activating factor （PAF） in endothelial cell. Methods： The patients with coronary heart disease and diabetes mellitus were divided into group A （without atorvastatin 20 mgqd）, group B （routine oral administration atorvastatin 20 mgqd 〉7 days）, group C （Routine oral administration atorvastatin 20mgqd 〉7days and Yishen Huoxue Jiedu decoction 150ml bid） ,15 people in each groups. Fasting blood samples were taken from three groups of patients in the morning to detect the liver and kidney function, blood glucose, blood lipid, coagulation function and other related indicators. Interleukin-6（IL-6）, interleukin-1β （IL-1β） and tumor necrosis factor （TNF- ct） in the mononuclear cells were detected by enzyme-linked immunosorbent assay （ELISA） And TNF-α protein expression levels was detected. Group A and C were treated with Yishen Huoxue Jiedu decoction 150ml bid after blood testing. Three groups of consecutive oral administration again after 4 weeks to check the same indicators. Results： The levels of TNF- α, IL-1β and IL-6 protein in group A and B were significantly higher than those in group C （P 〈0：05）. Conclusion： Yishen Huoxue Jiedu decoction can inhibit the expression of IL-6, IL-1β and TNF-α in monocytes induced by PAF. Yishen Huoxue Jiedu decoction combined with atorvastatin can inhibit PAF-induced expression of inflammatory mediators in endothelial cells.
