冠心止痛胶囊对内皮细胞保护作用时效关系研究

Research of the Time-protective Effect Relationship of Guanxin Zhitong Capsule on Human Umbilical Vein Endothelial Cells

目的:观察冠心止痛胶囊含药血清对内皮细胞保护作用的时效关系。方法:建立体外培养的人脐静脉内皮细胞H_2O_2氧化损伤模型,以冠心止痛胶囊不同采血时间所制备的含药血清对氧化损伤内皮细胞存活率、乳酸脱氢酶活性、一氧化氮、内皮素-1、组织型纤溶酶原激活物、组织型纤溶酶原激活物抑制剂的含量为指标,评价不同方案制备的含药血清对内皮细胞的保护作用。结果:不同采血时间所制备冠心止痛胶囊含药血清对H_2O_2诱导的人脐静脉内皮细胞损伤均具有保护作用,给药后2h、3h制备的含药血清的药效明显强于给药后1h制备的含药血清,给药后6h制备的含药血清药效弱于给药后1h制备的含药血清,但2组无统计学差异。结论:冠心止痛胶囊含药血清的药效与时间存在一定时效关系,以给药后2-3h所制备的含药血清药理效应最佳。

Objective： To study the time-protective effect relationship of Guanxin Zhitong capsule on human umbilical vein endothelial ceils. Methods： H2O2 oxidative injury model of human umbilical vein endothelial ceils （HUVECs） cultured in vitro was established. The effect of drug serum at different time after used Guanxin Zhitong capsule on oxidative damaged plasminogen activator, endothelial cell viability, lactate dehydrogenase activity, nitric oxide, endothelin-1, tissue-type tissue-type plasminogen activator inhibitor content were used to evaluated the time-protective effect relationship on HUVECs. Results： The serum containing Guanxin Zhitong capsule prepared by different blood collection time had protective effect on the injury of human umbilical vein endothelial cells induced by H202. The drug serum prepared at 2 h and 3 h after administration was significantly stronger than that at 1 h . The serum containing medicine prepared at the 6th hour after drug administration was weaker than the serum containing medicine prepared at 1 hour after administration, but there was no significant difference between the two groups. Conclusion： The pharmacological effect of Guanxin Zhitong capsule has a certain time-effect relationship with the time, and the pharmacological effect of the serum containing the drug serum is best after 2-3h.
