摘要
目的:探讨甘露糖结合凝聚素( mannose-binding lectin,MBL )基因多态性与青海地区藏族肺结核病易感性的相关性。方法以青海地区藏族肺结核患者(n=99)为实验组,青海地区藏族健康者(n=89)为对照组。启动区-550H/L(rs11003125)、+4P/Q(rs7095891)、-221X/Y(rs7096206)以及结构区的外显子1密码子52的 D ( rs5030737)、密码子54的 B ( rs1800450)、密码子57的 C (1800451),通过TaqMan探针实时定量聚合酶链反应(TaqMan探针法)检测出各位点的基因型。应用SPSS20.0统计软件,组间比较采用χ^2检验,检验水准α=0.05。结果通过TaqMan探针法检测MBL基因的6个SNP位点中rs1800450、rs7095891和rs7096206位点两组间基因型比较差异有统计学意义(P〈0.05);rs1800451和rsrs11003125位点两组间基因型比较无差异(P〉0.05)。结论 MBL基因的rs1800450、rs7096206和rs7095891位点多态性可能与青海地区藏族肺结核病易感性相关。
Objective To investigate the correlations between the mannose-binding lectin ( MBL) gene polymorphisms and the susceptibility to tuberculosis in Tibetan population in Qinghai province. Meth-ods Ninety-nine Tibetan patients with pulmonary tuberculosis in Qinghai province were recruited as the ex-perimental group and eighty-nine healthy Tibetan subjects were recruited as the control group. The genotypes of 6 single nucleotide polymorphisms ( SNPs ) in MBL gene including the start zone-550-H/L (rs11003125), +4P/Q (rs7095891) and -221X/Y (rs7096206) as well as the exon 1 codon 52 (allele D) (rs5030737), codon 54 (allele B) (rs1800450) and codon 57 (allele C) (rs1800451) were analyzed by using TaqMan probe real-time quantitative polymerase chain reaction ( TaqMan probe method ) . SPSS20. 0 software was used to comparatively analyze the differences in the genotypes and haplotypes of MBL gene between the two groups. Results The allele frequency in rs5030737 was not in accordance with Har-dy-Weinberg equilibrium. Significant differences in the genotypes between the two groups were observed in rs1800450, rs7095891 and rs7096206 (P〈0. 05), but not in rs1800451 and rs11003125 (P〉0. 05). Con-clusion The MBL gene polymorphisms in rs1800450, rs7096206 and rs7095891 sites were associated with the susceptibility to tuberculosis in Tibetan population in Qinghai province.
