摘要
以蒙古黄芪[Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao]种子萌发后的上胚轴、下胚轴和子叶为材料,采用MS培养基附加不同浓度植物生长调节剂进行愈伤组织的诱导,并诱导不定芽的发生,生根后得到完整的再生植株。在该过程中,采用正交试验分析6-BA、NAA、2,4-D、固化剂等条件对蒙古黄芪愈伤组织诱导和不定芽分化的影响。结果表明,下胚轴为诱导愈伤组织的最佳外植体取材部位,最佳愈伤组织诱导培养基为MS+1.0 mg/L 6-BA+0.2 mg/L 2,4-D,最佳的不定芽分化培养基为MS+1.0 mg/L 6-BA+0.1 mg/L NAA,而固化剂采用0.1%Gelrite为最佳。
With the epicotyls, hypocotyls and cotyledons from seed germination of Astragalus membranaceus(Fisch.)Bge. var.mongholicus(Bge.) Hsiao as explants, MS medium plus different concentrations of plant growth regulators was used to callus inducement and adventitious buds generation for plant regeneration. In this process, orthogonal experiment were used to analysis the impact of 6-BA,NAA,2,4-D and curing agent to callus inducement and adventitious buds differenciation of Astragalus membranaceus. The results showed that hypocotyl was the best site for explant callus inducement. The optimal medium for callus inducement was MS+1.0 mg / L 6-BA+0.2 mg / L 2,4-D and for adventitious buds differenciation was MS +1.0 mg / L 6-BA+0.1 mg / L NAA. The optimal curing agent was 0.1% Gelrite.
