组培过程中兜唇石斛的耐盐性研究

The Study on Salt Resistance in the Tissue Culture of Dendrobium aphyllum

以兜唇石斛试管苗为材料,研究组织培养过程中试管苗的生长状况,通过测定组织培养过程中分化苗合成的脯氨酸含量来确定兜唇石斛的耐盐性。最终得到最适合兜唇石斛试管苗生长的培养基是MS+20 g/L蔗糖+6.5 g/L琼脂+2 mol/L 6-BA+0.6 mol/L NAA;进行耐盐培养时得到脯氨酸含量最高的培养基是MS+20 g/L蔗糖+6.5 g/L琼脂+2 mol/L 6-BA+0.6 mol/L NAA+0.6%Na Cl。

The seedlings of Dendrobium aphyllum were used as materials to study the propagation and salt tolerance in the tissue culture. The content of the proline from seedlings could determine salt tolerance of Dendrobium aphyllum. Finally,the most suitable medium for Dendrobium aphyllum seedlings to grow was： MS＋20 g/L D（＋）-Sucrose＋6.5 g/L Agar＋2 mol/L 6-BA＋0.6 mol/L NAA.
