响应面法优化超声辅助提取荞麦中芦丁的工艺 被引量:3

摘要 为优化超声辅助乙醇浸提荞麦(Fagopyrum esculentum)中芦丁的工艺条件,利用响应面法优化荞麦芦丁提取工艺,以芦丁提取量为指标,通过单因素试验和响应面试验探讨超声时间、超声温度、液料比和乙醇体积分数对提取量的影响。结果表明,获得的最佳工艺为超声时间23 min、超声温度79℃、液料比30∶1(m L∶g)、乙醇体积分数90%。交互作用影响显著的因素为超声时间与乙醇体积分数、超声温度与乙醇体积分数。在此条件下进行验证,荞麦芦丁的提取量为0.416 1 mg/g,与模型预测值0.428 8 mg/g基本相符。模型可以较好地预测荞麦芦丁的提取量,响应面法对荞麦芦丁提取条件参数优化具有可行性。 In order to optimize the technological condition of ultrasonic-assisted ethanol extraction of rutin from Fagopyrum esculentum, taking the yield of rutin as the index, The effects of four main variables, i.e., ultrasound time, ultrasound temperature, liquid-solid ratio and percentage of ethanol concentration on the extraction yeild of rutin were discussed by single-factor experiments, the optimal process of extracting rutin from Fagopyrum esculentum were obtained by response surface methodology. The results showed that, the optimum extraction conditions were as follows: extraction time of 23 mins, extraction temperature of 79 ℃, liquid-solid ratio of 30∶1 and ethanol concentration of 90%. The interactions were prominent between ultrasound time and ethanol concentration, also between extraction temperature and ethanol concentration. Under these conditions, a mean value of 0.416 1 mg/g(n=3) was observed, which was well matched with the predicted value of 0.428 8mg/g. The proposed prediction model could accurately predict the extraction yield of rutin, which suggested it was feasible to optimize the extraction conditions of rutin from Fagopyrum esculentum using response surface methodology.
