补肾养血方药对气态甲醛慢性染毒小鼠子宫内膜容受性的影响

摘要 目的:探讨补肾养血方药对气态甲醛慢性染毒小鼠子宫内膜容受性的影响。方法:将30只性成熟SPF级雌性昆明种小鼠随机分为空白对照组、染毒组和中药治疗组,每组10只,空白对照组给予吸入经过滤的新鲜空气,染毒组和中药治疗组给予1.0 mg/m3浓度气态甲醛动态吸入式染毒,6 h/d,同时中药治疗组每日予补肾养血中药灌胃,空白对照组和染毒组予等量生理盐水灌胃,连续14 d;将小鼠雌雄(2:1)合笼处理,于孕5 d处死小鼠,取出子宫,计算子宫脏器系数,观察胚胎着床情况;用RT-PCR和Real-time PCR法检测各组小鼠子宫Hoxa10、Hoxa11 m RNA的表达。结果:染毒组小鼠子宫脏器系数、子宫Hoxa10、Hoxa11 m RNA表达均低于空白对照组,差异均有统计学意义(P<0.05或P<0.01),胚胎着床数低于空白对照组,但差异无统计学意义(P>0.05)。中药治疗组小鼠子宫Hoxa10、Hoxa11 m RNA表达高于染毒组,差异均有统计学意义(P<0.05或P<0.01)。结论:补肾养血方药能使气态甲醛慢性染毒小鼠子宫Hoxa10、Hoxa11 m RNA表达升高,从而改善子宫内膜容受性。 Objective: To study the influences of Traditional Chinese Herbs with tonitying kidney and nourishing blood effect on the endometrial receptivity of mice chronically exposed to gaseous formaldehyde. Methods: The 30 sexually mature Kunming female mice of SPF level were randomly assigned into control group, chronic gaseous formaldehyde exposure group and treatment group, ten mice in each. The mice were given filtered fresh air in control group and gaseous formaldehyde for dynamic inhalation, 1.0 mg/m3, 6 t/d, in exposure group and treatment group. Simultaneously, the mice were given decoction of Traditional Chinese herbs with tonifying kidney and nourishing blood in treatment group, while the same amount of physiological saline in the control group and exposure group, for 14 days. Then, the female and male mice were co-caged with ratio of 2:1. The mice were killed at the 5th day after pregnancy, and the uterus was taken out; The organic coefficient of uterus was calculated and the embryonic implantation situation was observed. The expression levels of Hoxa10 and Hoxall mRNA in uterus tissues were detected by means of RT-PCR and Real-time PCR. Results: The organic coefficient and the expression levels of Hoxal0 and Hoxall of uterus in exposure group mice were decreased compared with the control group, and there were statistically significant differences (P〈0.05 or P〈0.01). The numbers of embryonic implantation had decreasing trend, but there was no statistically significant difference (P〉0.05). The expression levels of Hoxa10 and Hoxall in the treatment group were increased compared with the exposure group and there was statistically significant difference (P〈0.05 or P〈0.01). Conclusion: Traditional Chinese herbs with tonifying kidney and nourishing blood function could increase the expressions of Hoxa10 and Hoxall mRNA in the uterus of mice chronically exposed to gaseous formaldehyde, thereby improving the endometrial receptivity.
