摘要
于2014年11月和2015年3月从天津辖区分别采集了淡水水样和海水水样,分别利用BG11和f/2培养基,采用平板分离法对水样中的微藻进行了分离。将分离得到的菌株扩大培养后提取基因组DNA,经聚合酶链式反应(PCR)扩增18S r DNA,将PCR产物测序后与NCBI基因数据库比对确定品种。试验分离得到菌株179株,其中有143株经序列比对,确定分属于绿藻门6个不同的属,建立了系统发育树,同时初步建立了天津水域冬、春两季微藻种质资源库。
Water samples were collected from both freshwater and seawater in Tianjin in Novermber 2014 and March 2015.Microalgae strains in the water samples were isolated by BG11 or f/2 media. Genomic DNA of each strain was extracted after cultivation and 18S rDNA was amplified by PCR and sequenced. By blasting the sequencing results in the NCBI Gen Bank database,the genus of those microalgae were identified. A total of 179 strains were isolated,wherein 143 fell into six genera of Chlorophyta. A phylogenetic tree was built based on the sequencing results. Moreover,a preliminary collection of native microalgae in Tianjin was established.
