脂肪干细胞覆膜聚丙烯补片在大鼠体内的生物相容性 被引量:6

Biocompatibility of polypropylene mesh scaffold with adipose-derived stem cells in rats

摘要 目的探讨脂肪干细胞(ADSCs)覆膜聚丙烯补片在大鼠体内的生物相容性。方法培养扩增检测大鼠脂肪干细胞后,将其接种到聚丙烯补片上,体外培养10 d。成年雄性SD大鼠12只,个体质量约(250±20)g,随机分为覆膜补片组和未覆膜补片组,每组6只。分别将聚丙烯补片和ADSCs覆膜聚丙烯补片植入大鼠的腹壁肌肉表面,30 d后观察补片侵蚀、粘连情况,测量补片厚度、硬度、韧性等,并进行组织学观察。结果细胞呈长梭形,并形成小集落,呈束状、菊花状或旋涡状生长,活力良好,免疫荧光检测结果显示干细胞特异性标记CD90表达≥97%。ADSCs在聚丙烯补片上附着生长良好。植入大鼠体内30 d后,ADSCs覆膜聚丙烯补片炎症反应较聚丙烯补片轻,炎症细胞评分[(0.95±0.23)分vs(2.62±0.40)分,P<0.05],而成纤维细胞增殖和新生血管生成却更多,分别为[(2.6±0.4)分vs(1.8±0.4)分,P<0.05]和[(2.4±0.4)分vs(1.5±0.4)分,P<0.05]。30 d后ADSCs覆膜聚丙烯补片的硬度与聚丙烯补片无明显差别[(2.1±0.3)分vs(2.2±0.3)分,P>0.05],而厚度[(2.67±0.37)分vs(1.13±0.24)分,P<0.05]、韧性[(2.67±0.31)分vs(2.27±0.26)分,P<0.05]等均大于聚丙烯补片。结论 ADSCs覆膜聚丙烯补片相较聚丙烯补片植入大鼠体内后可以减轻局部炎症反应、增加局部腹壁顺应性。 Objective To investigate vivo biocompatibility of polypropylene mesh scaffold with adipose-derived stem cells (ADSCs) in rats. Methods ASDCs were seeded onto polypropylene mesh scaffold after passage and amplification, and were cultured in vitro for 10 days. 12 SD male rats [ weight (250 ± 20) g], were randomly divided into two groups, including 6 rates in each group. The polypropylene mesh with and without ADSCs were separately implanted into the surface of abdominal wall muscle. Adhesion and erosion of the mesh were evaluated, and thickness, solidity and tenacity were measured after 30 days, as well as historical observation. Results ADSCs showed good adhesion and growth on polypropylene mesh with long spindle cell shape, and grew in bundle, or in chrysanthemum or spiral shape with good vitality. 30 days after implantation into rats in vivo, polypropylene mesh with ADSCs had less inflammatory reaction than that without ADSCs, with inflammatory cell score [ (0.95 ±0.23) vs (2.62 ±0.40), P 〈0.05], but more fibroblast proliferation [ (2.6 ± 0.4 ) vs ( 1.8 ± 0.4 ), P 〈 0.05 ] and new blood vessel formation [ ( 2.4 ±0. 4) vs ( 1.5 ± 0.4), P 〈 0.05 ]. There was no significant difference in solidity [ ( 2.1 ± 0.3 ) vs ( 2.2 ± 0.3 ), P 〉 0.05 ], while polypropylene mesh with ADSCs had increased thickness [ (2.17 ± 0.37 ) vs ( 1.13 ± 0.24), P 〈 0.05 ] and tenacity [ (2.67± 0.31 ) vs ( 2.27 ± 0.26), P 〈 0.05 ] than that without ADSCs. Conclusion Polypropylene mesh with ADSCs has less inflammatory reaction and better compliance of the abdominal wall after implantation in rats in vivo than that without ADSCs.
