摘要
目的探讨四君子汤保护骨损伤小鼠细菌易位及对骨修复的促进作用。方法在100只健康小鼠中选取10只为对照组,其余90只小鼠造模后随机分为中药组、抗生素组、自然恢复组(各30只)。抗生素组小鼠每天腹腔内注射青霉素和链霉素,中药组在此基础上给予四君子汤灌胃,自然恢复组以0.9%Na Cl溶液灌胃。造模10天后,检测肝脏细菌易位及巨噬细胞吞噬功能;观察中药组、抗生素组和自然恢复组骨折后2周、4周、6周和8周骨痂面积及骨体积分数(BVF)。结果中药组小鼠肝脏细菌易位率和肝脏活菌计数均显著低于自然恢复组和抗生素组(P<0.01);中药组巨噬细胞吞噬能力显著高于自然恢复组和抗生素组(P<0.01);中药组骨痂面积缩小最快(P<0.01);在不同时间点,中药组BVF均高于自然恢复组和抗生素组(P<0.01)。结论四君子汤能够抑制骨损伤小鼠细菌易位、提高巨噬细胞的吞噬功能以促进骨折修复。
Objective To discuss the effects of Sijunzi Decoction on suppressing bacterial translocation and promoting bone repair in mice with bone injury.Methods Ten mice were taken as the control group from 100 healthy mice. After modeling the other 90 mice were randomly divided into the traditional Chinese medicine( TCM) group,antibiotic group and recovery group,30 mice in each group. The antibiotic group was treated with penicillin and streptomycin by intraperitoneal injection,the TCM group was treated with Sijunzi Decoction based on the treatment for the antibiotic group,and the recovery group was treated with0. 9% normal saline by intragastric administration. 10 days after modeling,the bacterial translocation in the liver and the phagocytic function of macrophages were detected. The scar areas and bone volume fraction( BVF) of the TCM group,antibiotic group and recovery group were checked after 2 weeks,4 weeks,6 weeks and 8 weeks. Results The rate of bacterial translocation and liver living bacteria count in the TCM group were significantly lower than those in the recovery group and antibiotic group( P〈0. 01); the phagocytic capacity of macrophages in the TCM group was significantly higher than that in the recovery group and antibiotic group( P〈0. 01); the scar areas reduced most quickly in the TCM group( P〈0. 01); the BVF in the TCM group at different time points were all higher than that in the recovery group and antibiotic group( P〈0. 01). Conclusion Sijunzi Decoction can inhibit the bacterial translocation and improve the phagocytic function of macrophages to promote bone repair in bone damage mice.
