扶正康肺方对肺癌患者肺泡巨噬细胞胞浆游离钙浓度及其生成细胞因子的影响

摘要 目的探讨扶正康肺方对肺癌患者肺泡巨噬细胞胞浆(alveolarmacrophage,AM)游离钙浓度及生成的细胞单核细胞趋化蛋白(MCP-1)、白介素-12(IL-12)、一氧化氮(NO)分子的影响.方法采用支气管肺泡灌洗、细胞培养和荧光指示剂方法,并分别用干扰素-γ(IFN-γ)、扶正康肺方干预AM培养,检测30例肺癌患者AM钙浓度及生成的MCP-1、IL-12和NO浓度.结果①肺癌患者AM基础制剂胞内钙浓度;并产生一定量的MCP-1、IL-12和NO;肺癌患者AM胞内钙浓度是高于正常组(P〈0.05);②但是MCP-1、IL-12和NO均低于正常对照组的(P〈0.05).③加入大剂量扶正康肺方后加入LPS的情况下与同组内仅加入LPS相比,胞内游离钙浓度明显升高(P〈0.05),两组间比较差异统计学意义(p〉0.05).加入小剂量扶正康肺方后加入LPS的情况下与同组内仅加入LPS相比,胞内游离钙浓度升高,但是暂时未有统计学意义(P〈0.05).④IFN-γ、扶正康肺方干预下AM胞内钙浓度、MCP-1、IL-12和NO的浓度较干预前明显升高(P〈0.05).扶正康肺方大剂量刺激下刺激下,MCP-1、IL-12和NO与小剂量刺激相比较,是是升高的,但差异无统计学意义(P〉0.05).结论扶正康肺方能促进肺癌患者AM激活,促进MCP-1、IL-12和NO效应分子;能提高AM免疫活性. Objective To examine the effects of fuzhengkangfei on cytosolic free calcium in alveolar macrophages and on macrophages-derived MCP-1、IL-12 and NO. Methods Techniques of bronchoaiveolar lavage, cell culture-and fluorescent caleium labeling with Fura-2/ AM were emoloyed to assess the cytosolic levels of calcium and secretion of MCP-1、IL-12 and NO. Results ①[Ca2+]i were significantly increased in lung cancer,as compared with the control group (P〈0.05).②[Ca2+]i were further elevated after stimulation LPS (P〈0.05).③Addition of LPS after small dose fuzhengkangfei incubation shown to outcoming in higher level of f [Ca2+]i compared with addition of LPS alone; but wes not significantly . Addition of LPS after large dose fuzhengkangfei incubation shown to outcoming in higher significantly level of [Ca2+]i compared with addition of LPS alone. ④AM in all the lung cancer patients produced MCP-1、IL-12 and NO. The activity of MCP-1、IL-12 and NO was lower in the BALF and in the cultured supernatants of AM of lungs cancer than AM of control groups. The concentrations of MCP-1、IL-12 and NO in the cultured supernatants of AM cultivated with small dose or large dose fuzhengkangfei were higher than those in the nonirritant groups [(P〈0.05); but there were higher in large dosegroup than those in the small dose groups (P〈0.05) .Conclusion The fuzhengkangfei increased [Ca2+]i in lung cancer ; concentrations of MCP-1、IL-12 and NO in the cultured supernatants of AM with fuzhengkangfei were induced higher . (P 〈0 .05 ). Conclusion fuzhengkangfei improved immunocompetence of alveolar macrophages in lung cancer.
