组织块贴壁法培养和鉴定SD大鼠主动脉平滑肌细胞

目的研究 SD 大鼠主动脉平滑肌细胞的培养方法并鉴定,为动脉粥样硬化相关疾病的病因机制研究提供实验材料.方法应用组织块贴壁法培养SD 大鼠主动脉平滑肌细胞并进行传代.倒置相差显微镜下观察细胞形态及生长情况,并进行免疫细胞化学鉴定. 结果成功培养SD大鼠主动脉平滑肌细胞并传代.倒置相差显微镜下细胞生长呈典型“峰-谷”状.免疫细胞化学染色胞浆内平滑肌肌动蛋白均呈阳性表达.结论组织块贴壁法培养SD大鼠主动脉平滑肌细胞,此方法操作简便,可短期获得纯度较高的血管平滑肌细胞,可作为研究动脉粥样硬化相关疾病良好的体外实验模型.

Objective To study the method for primary culture of rabbit arterial smooth muscle cells.To provide experimental material for the cause and mechanism research atherosclerosis related disease. Methods SD rat arterial smooth muscle cells were cultured with tissue explants adherent method and could be passaged. The observation of cell morphology and growth situation were observed under the inverted phase contrast microscope and were identified with the method of immunocytochemistry. Results SD rat arterial smooth muscle cells were cultured successfully and could be passaged.Under inverted phase contrast microscope,the growth of the cells showed a typical peak-and-vally pattern. The cytoplasm smooth muscle actin were positive with immunocytochemistry staining.Conclusions The culture techenique of tissue explants adherent method has the advantages of easy to operate,high purity of vascular smooth muscle cells in a shorter cycle time.They are good vitro model of Atherosclerosis related diseases
