摘要
目的:通过检测HBx 和Beclin 1在原发性肝癌、癌旁组织的表达水平,结合临床病理指标,探讨HBx 和Beclin 1在HCC 的发病机制中作用及其相互关系.方法:收集本院2014年1月 -2015年12 月由病理确诊的 HCC 患者64例,取其肝癌组织及其癌旁组织(距肿瘤边缘〉2cm),患者术前未经任何抗肿瘤治疗,用免疫组化、Western Blot 及Real Time PCR 方法检测 HBx 及 Beclin l 在上述标本中的表达情况,并将实验结果与患者的临床资料结合进行统计学分析.结果 :HBx 在肝癌组织表达高于癌旁组织:免疫组化、Western Blot、Real Time PCR 技术均检测肝癌组织中 HBx 表达量高于癌旁组织,差异具有统计学意义(P〈0.05).Beclin1在肝癌组织中表达高于癌旁组织:免疫组化、Western Blot、Real Time PCR 技术均检测肝癌组织中Beclin1表达量高于癌旁组织,差异具有统计学意义(P〈0.05).且原发性肝癌中 HBx 与 Beclin 1 表达成正相关.结论 :HBx 和 Beclin 1 在HCC 中高表达,两者相互作用且共同参与HCC 的发生、发展过程,联合检测 HBx 和 Beclin 1 对判断HCC 的发生和转移有重要价值,是有潜力的肝癌治疗的候选靶基因.
Objective To detect HBx and Beclin 1 expression in Hepatocellular carcinoma (HCC) and study their correlations to the clinic pathological features. To explore the effect of HBx and Beclin1 expression in the occurrence ,development and metastasis of HCC and their interrelation. Methods We had collected 64 cases of HCC tissues and their surrounding tissues which had been diagnosis by Dr. of the department of 3201th Affiliated Hospital of Xi'an Jiaotong University from 2014,01--2015,12. We examed the expression of mRNA and protein of the HBx and Beclin l by Western Blot , immunohistochemistry (IHC) and Real-Time PCR.The outcoming were used to analyze the relationship between the expression of HBx and Beclin 1 mRNA and protein in HCC and the information of pathology and clinic. Results: HBx expressed higher in HCC than their surrounding tissues. The IHC consequence of the expression of the HBx protein in HCC were higher than their surrounding tissues;HBx protein expression in HCC tissues were higher than the HCC surrounding tissues in Western Blot; HBx mRNA expression in HCC tissues were higher than in HCC surrounding tissues by Real Time PCR. Beclin 1 expressed higher in HCC than their surrounding tissues.The IHC consequence of the expression of the Beclin1 protein in HCC were higher than their surrounding tissues. Beclin1 protein expression in HCC tissues were higher than the HCC surrounding tissues in Western Blot; Beclin1 mRNA expression in HCC tissues were higher than in the HCC surrounding tissues by Real Time PCR. The expression rate of HBx was significantly correlated with that of Beclin 1 in HCC. Conclusion: There were higher expression of HBx and Beclin 1 in HCC. HBx and Beclin 1 interact one another and participate in the occurrence、development and metastasis of HCC. Combined detection of HBx and Beclin 1 has important value for prediction of occurrence and metastasis. They are both potential target gene for the therapy of HCC.
