南葶苈子提取液对心力衰竭大鼠心功能的影响 被引量:23

摘要 目的探讨南葶苈子提取液对心力衰竭大鼠心功能的影响。方法将10只SD大鼠作为假手术组,40只SD大鼠采用肾上腹主动脉缩窄术制作大鼠心力衰竭模型,随机分为模型组、醇提取组、水提取组及地高辛组。假手术组及模型组给予5ml/(kg·d)生理盐水灌胃,醇提取组予南葶苈子醇提液10g/(kg·d)灌胃,水提取组予南葶苈子水提液10g/(kg·d)灌胃,地高辛组予地高辛片0.05mg/(kg·d)灌胃,4周后观测其血流动力学、心肌病理组织切片、超声心动图变化。结果病理学观察显示,模型组心肌细胞坏死严重,心肌纤维排列紊乱;与模型组比较,醇提取组、水提取组及地高辛组心肌细胞坏死轻微,心肌纤维排列尚可。血流动力学检测显示,模型组左心室收缩压(LVSP)下降,左心室舒张末压(LVEDP)升高,左心室内压最大上升速率(+dp/dtmax)及左心室内压最大下降速率(-dp/dtmax)下降,比较有统计学意义(P<0.05);与模型组比较,醇提取组、水提取组及地高辛组LVSP升高,LVEDP降低,+dp/dtmax升高,比较有统计学意义(P<0.05)。超声心动图结果显示,模型组左心室舒张末期内径(LVIDd)、左心室收缩末期内径(LVIDs)升高,射血分数(EF)、短轴缩短率(FS)降低,比较差异有统计学意义(P<0.05);与模型组比较,醇提取组、水提取组及地高辛组LVIDd、LVIDs降低,EF、FS升高,比较差异有统计学意义(P<0.05)。结论南葶苈子提取液可改善心力衰竭SD大鼠心功能各项指标,从而增强心功能。 Objective To study the effects of descurainiae extract on cardiac function in heart failure rats .Methods Ten SD rats were selected as the sham operation group .After operated by suprarenal abdominal aortic coarctation ,forty SD rats were randomly divided into model group ,ethanol extract group ,water extract group and digoxin group .Rats in the sham operation group and the model group were both given normal saline 5ml/(kg·d) by gavage .Rats in the ethanol extract group ,the water extract group and the digox-in group were treated respectively with ethanol descurainiae extract 10g/(kg·d) ,water descurainiae extract 10g/(kg·d) ,digoxin 0 . 05mg/(kg·d) by gavage .After four weeks ,the hemodynamics ,myocardial pathological section and ultrasonic cardiogram of the rats were observed. Results Pathology observation showed that there were serious myocardial necrosis and disordered myocardial fibers in the model group while slight myocardial necrosis and relatively regular myocardial fibers in the ethanol extract group ,the water extract group and the digoxin group .Hemodynamics examination showed that the LVSP was decreased ,LVEDP was increased and the maximum left ventricular pressure rising and dropping rates ( +dp/dtmax) were reduced in the model group with statistical significance (P〈 0 .05) .Compared with the model group ,the LVSP was increased ,LVEDP was decreased and the maximum left ventricular pressure rising rate (+ dp/dtmax) was increased in ethanol extract group ,the water extract group and the digoxin group with statistical significance (P〈 0 .05) .Ultrasonic cardiogram examination showed that LVIDd and LVIDs was significantly increased ,EF and FS was significantly decreased in the model group (P〈 0 .05) .Compared with the model group ,LVIDd and LVIDs was significantly decreased ,EF and FS was significantly increased in ethanol extract group ,the water extract group and the digoxin group (P〈 0 .05) . Conclusions Descurainiae extract could effectively enhance cardiac function of SD rats with heart failure since all the indicators were improved .
