SUMO蛋白酶1对慢病毒感染骨髓瘤细胞生长的影响

摘要 目的用含有SUMO蛋白酶1干涉载体的shRNA慢病毒感染骨髓瘤细胞系XG-7,探讨SUMO蛋白酶1对骨髓瘤细胞的影响作用,为临床治疗提供参考依据。方法用含有SUMO蛋白酶1干涉载体的shRNA慢病毒和con慢病毒感染骨髓瘤细胞系XG-7,流式细胞仪和荧光显微镜观察骨髓瘤细胞系XG-7慢病毒感染效率,Q-PCR和western blot检测XG-7骨髓瘤细胞系感染shRNA和con病毒后SUMO蛋白酶1的敲除效率。结果con病毒和shRNA病毒感染XG-7骨髓瘤细胞系48h后,病毒感染率均>98.00%;shRNA组SUMO蛋白酶1RNA和蛋白表达量、骨髓瘤细胞系XG-7存活细胞比例均显著低于con组(P<0.05)。结论 SUMO蛋白酶1敲除能够抑制骨髓瘤细胞增殖,促进凋亡,影响骨髓瘤细胞的细胞周期和集落形成。 OBJECTIVE ShRNA lentivirus containing SUMO protease 1interferometer vector was used to infect myeloma cell line XG-7,so as to explore the influence of SUMO protease 1on myeloma cells,and provide references for clinical treatment.METHODS ShRNA lentivirus containing SUMO protease 1interferometer vector and con lentivirus were used to infect myeloma cell line XG-7,flow cytometry and fluorescence microscopy were used to observe myeloma cell line XG-7lentivirus infection efficiency,Q-PCR and Western blot were used to detect the knockout efficiency of SUMO protease 1in myeloma cell line XG-7infected with shRNA virus and con virus.RESULTS The infection rates of XG-7myeloma cell lines at 48 hafter con virus and shRNA virus infection were above 98%.RNA and protein expression levels of SUMO protease 1and the myeloma cell line XG-7cell survival rate of shRNA group were less than those of con group(P〈0.05).CONCLUSIONKnockout of SUMO protease 1can inhibit myeloma cell proliferation,promote apoptosis,and influence cell cycle and colony formation of myeloma cells.
