口腔种植体周围炎对颌骨成骨细胞的增殖分化能力的影响研究

Effects of peri-implantitis on proliferation and differentiation of mandibular osteoblasts

目的探讨口腔种植体周围炎对颌骨成骨细胞增殖分化能力的影响,为临床治疗提供参考。方法收集医院口腔科松动并拔出种植体患者7例作为观察组,拔出第三磨牙身体健康者7例作为对照组,取两组患者的颌骨组织进行成骨细胞的分离培养和纯化,MMT测定成骨细胞增殖能力,RT-PCR法测定成骨细胞Ⅰ型胶原(Ⅰcollagen,Col I)mRNA水平,Western blot检测成骨细胞Col I蛋白水平。结果第7天时,观察组成骨细胞OD值低于对照组(P<0.05),第3天时,观察组成骨细胞OD值低于对照组,但差异无统计学意义;观察组成骨细胞Col I蛋白相对表达量明显低于对照组(P<0.05)。结论口腔种植体周围炎能够抑制颌骨成骨细胞的增殖和分化。

OBJECTIVE To explore effects of peri-implantitis on proliferation and differentiation of mandibular osteoblasts,so as to provide references for clinical tretment.METHODS Seven cases of patients with implant loosening and were pulled out were collected as observation group,and 7cases of healthy pulled out third molars as control group.The jaw bone tissue of the two groups of patients were taken for isolation,culture and purification of osteoblasts.MMT was used to assay osteoblasts proliferation,RT-PCR was used to assay osteoblasts Ⅰ collagen（ColⅠ）mRNA level,and Western blot was used to detect Col I protein level of osteoblasts.RESULTS The OD value of osteoblasts of observation group was lower than that of control group at the 7th day（P〈0.05）,the OD value of osteoblasts of observation group was lower than that of control group at the 3rd day,but the difference was not significant.The expression of ColⅠ protein of osteoblasts of observation group was significantly lower than that of control group（P〈0.05）.CONCLUSIONPeri-implantitis can inhibit the proliferation and differentiation of the mandibular osteoblasts.
