摘要
以新羽化的光肩星天牛(Anoplophora glabripennis)雄成虫触角为材料,采用RNA转录5'末端转换(SMART)技术构建全长c DNA文库。经验证,该文库容量为1.35×106pfu/m L,重组率为95%。从中随机挑取48个克隆进行表达序列标签(ESTs)测序,有效序列为45条,其中35条序列有相关同源信息,24条为全长序列,完整性比率为68.6%。
The full length cDNA library of Anoplophora glabripennis antenna was constructed by SMART RNA ( switching mecha-nism at 5′end of RNA transcript) which consisted of 1.35×10^6 pfu/mL and the recombinant percentage was 95%.Forty-eight clones were randomly selected from the cDNA library , the ESTs ( expressed sequence tags ) were sequenced .Forty-five were effective , 35 homologous sequences which contained 24 full-length sequences were found by blasted in the NCBI nonredundant nucleotide databases , and the integrity ratio of the full-length sequences was 68.6%.
