摘要
选取5种破壁灵芝孢子粉为研究对象,使用紫外分光光度法,建立了一种快速、简单、高灵敏度、环保,专门针对灵芝孢子粉中多糖测定的方法。样品以纯水恒温超声提取,经苯酚-硫酸法显色后,在紫外波长490 nm处测定吸光度。葡萄糖的质量浓度在0.1~0.8 mg/mL范围内呈良好的线性关系(r=0.999 4),方法的检出限为0.043 mg/mL,方法精密度(RSD)为0.23%,回收率为86.4%~92.8%,完全能够满足于破壁灵芝孢子粉中多糖成分的分析研究。
5 kinds of broken cellular wall of Ganoderma lucidum spore were chosen as study subjects. An analytical method was developed to determine polysaccharide in Ganoderma lucidum spore by ultraviolet spectrometry. This method was rapid, simple, sensitive and amity-environment. The samples were extracted with pure water by ultrasonic, colored by phenolsulfuric acid method. The absorbance was detected at UV wavelength 490 nm. The results indicated that the standard curves of glucose showed good linearity in the range ration of 0.1-0.8 mg/mL (r=0.999 4), LOD was 0.043 mg/mL, and the method precision (RSD) was 0.23%. The recoveries of polysaccharide ranged from 86.4% to 92.8%. The proposed method could be used for the determination of polysaccharide in broken cellular wall of Ganoderma lucidum spore.
