摘要 本文建立检测黄颡鱼Pelteobagrus fulvidraco肌肉中叶黄素含量的反相超高效液相色谱分析方法。样品经95%乙醇提取,ACQUITY UPLC BEH C_18色谱柱(2.1mm×100mm,1.7μm)分离,以89%甲醇-1%乙腈-10%水为流动相等度洗脱,二极管阵列检测器检测,检测波长446nm,外标法定量。结果显示:叶黄素在0.04~0.8μg·mL^(-1)范围内线性良好,此方法检出时限0.03mg·kg^(-1)。添加量在0.5mg·kg^(-1)、1.0mg·kg^(-1)、5.0mg·kg^(-1)时,叶黄素的回收率为82.0%~84.2%,相对标准偏差在10%范围内。结果表明:该方法可用于黄颡鱼中叶黄素成分的定性和定量检测。 A reversed-phase ultra-performance liquid chromatography method was established for the determination of lutein in muscle ofPelteobagrus fulvidraeo. The sample was extracted by 95% alcohol. Chromatographic separation was performed at the conditions of ACQUITY UPLC BEH C18 column (2.1 mm x 100 mm, 1.7 μm) and 89% methcol-1% acetonitrile-10%water as the mobile phase with isocratic elution. It was detected on a photodiode array detector at the wavelength of 446 nm and quantified by external standard method. The results showed that the method had a good linear correlation within the range from 0.04μg·mL^-1 to 0.8 μg·mL^-1, and the detection limit was 0.03 mg·kg^-1. The average recoveries of lutein varied from 82.0% to 84.2% spiked at 0.5 mg·kg^-1, 1.0 mg·kg^-1 , 5.0 mg·kg^-1 with relative standard deviations less than 10%. This developed method is expeditious and reliable, especially feasible for quantitative and qualitative analysis of lutein in Pelteobagrus fulvidraeo.
