高效液相色谱法测定天眩四物汤中天麻素含量

Content Determination of Gastrodine in Tianxuan Siwu Decoction by HPLC

目的 建立测定天眩四物汤中天麻素含量的高效液相色谱（HPLC）法。方法 采用高效液相色谱法,色谱柱为伊利特Hypersil ODS2-C18柱（250 mm×4.6 mm,5μm）;流动相为乙腈（A）-0.1%磷酸（B）梯度洗脱,0-13.5 min,A为1.9%,14-23 min,A为40%;流速为1 mL/min;检测波长为220 nm;柱温为25℃;理论板数按天麻素峰计算应不低于1 500。结果 天麻素质量浓度在0.116-1.160μg/L范围内与峰面积积分值线性关系良好,平均回收率为100.00%,RSD=1.54%（n=6）。结论 该方法简便、快速、重复性好、专属性强,可为制订天眩四物汤质量控制标准和有效控制天眩四物汤的内在质量提供技术参考。

Objective To establish an HPLC method for the content determination of gastrodine in Tianxuan Siwu Decoction. Methods The HPLC method was used. The chromatographic column was the Ealet Hypersil ODS2- C18 column （250 mm×4. 6 mm,5μm）;acetonitrile （A） - 0. 1% phosphoric acid （B） was used as the mobile phase,gradient elution,0-13.5 min,A 1.9%, 14-23 min,A 40% ,the flow rate was 1 mL/min;the detection the wavelength is 220 nm,the column temperature was 25 ℃. The number of theoretical plates according to the calculation of the peak of the peak should not be less than 1 500. Results The gastrodin concentration was linear in the range of 0. 116-1. 160 μg/L, the average recovery rate was 100. 00%, RSD = 1.54% （n =6）. Conclusion The method is simple, rapid, reproducible and specific, can provide technical reference for the formulation of internal quality standards and Tianxuan Siwu Decoction.
