摘要
目的:观察平动汤及利他林对注意力缺陷多动障碍(attention deficit hyperactivity disorder,ADHD)模型大鼠下丘脑-垂体-肾上腺轴功能的影响。方法:将6只WKY大鼠设为正常组,30只自发性高血压WKY大鼠分为模型组,利他林组,平动汤低剂量组,平动汤中剂量组,平动汤高剂量组,灌胃4周后,用放射免疫法测定大鼠血清中促肾上腺皮质激素(adrenocorticotrophic hormone,ACTH)及皮质醇的水平。结果:模型组大鼠ACTH水平高于正常组,皮质醇水平低于正常组(P<0.05),提示模型复制成功。治疗后与模型组比较,平动汤中剂量组ACTH差异有统计学意义(P<0.05);与利他林组比较,平动汤中剂量组ACTH差异有统计学意义(P<0.05)。治疗后与模型组比较,平动汤中剂量组、利他林组大鼠血清皮质醇差异均有统计学意义(P<0.05)。结论:平动汤可能是通过降低ACTH水平,提高皮质醇水平来调节下丘脑-垂体-肾上腺轴功能,达到治疗ADHD的目的。
Objective: Explore the effects of Ping Dong Tang and Methlyphenidate( MPH) on the animal model of attention deficit hyperactivity disorder under hypothalamic-pituitary-adrenal axis function. Methods: Six WKY rats were normal group,thirty SHR rats were randomly divided into five groups: ADHD model group,MPH groups,Ping Dong Tang-low-dose groups,Ping Dong Tangmedium-dose groups,Ping Dong Tang-high-dose groups. All the rats were dosed for four weeks in a row,and then used radioimmunoassay to detect the adrenocorticotropic hormone and cortisol expression in the serum. Results: The level of adrenocorticotropic hormone in model groups was higher than normal,cortisol levels lower than the normal group. After treatment can be reduced adrenocorticotropic hormone levels,at the same time Ping Dong Tang groups-medium-dose groups had difference that was statistically significant( P〈0. 05). Treatment groups had a trend to raise serum cortisol level,Ping Dong Tang-low dose groups and MPH groups could improve the serum cortisol levels,the differences were statistically significant( P〈0. 05). Conclusions: Ping Dong Tang and MPH achieve the treatment of purpose of attention deficit hyperactivity disorder probably by reducing adrenocorticotropic hormone levels and improving the serum cortisol levels to regulate the hypothalamic-pituitary-adrenal axis.
