匹罗卡品颞叶癫痫大鼠海马结构及功能网络损伤的磁共振成像研究 被引量:2

An MRI study on abnormal hippocampus related structure and functional connectivity in pilocarpine-induced temporal lobe epilepsy rats

摘要 目的研究慢性期氯化锂一匹罗卡品(LIP)颞叶癫痫(TLE)大鼠的海马结构及功能网络损伤情况。方法30只sD大鼠随机分为TLE组(20只)和对照组(10只)。对TLE组大鼠进行LIP造模,对照组注射等量生理盐水。造模6周后,对大鼠在7.0T磁共振下进行结构像及功能像扫描,并比较两组大鼠海马灰质体积及以背侧海马为种子点的功能连接度。结果TLE组12只大鼠在造模后出现癫痫持续状态,6周后其中10只大鼠观察到自发性癫痫发作。结构磁共振数据分析,TLE组和对照组大鼠的海马灰质体积分别为(85.76±1.62)mm。和(102.51±1.97)mm2,组间比较差异有统计学意义(P〈0.05)。功能磁共振成像分析显示,TLE组大鼠与对照组相比,背侧海马与同侧腹侧海马、杏仁核功能联系减弱(P〈0.05),而与皮质联系增强(P〈o.05)。结论LIP-TLE模型大鼠在进入慢性期后,与对照组相比,出现海马体积减小的结构损伤,以及海马与边缘系统功能连接减弱、与皮质功能连接增强的功能网络异常。 Objective To study the abnormal hippocampal structural and functional connectivity in pilocarpine-induced temporal lobe epilepsy (TLE) rats. Methods Thirty SD rats were randomized into TLE group ( n = 20) and control group ( n = 10 ). Lithium-pilocarpine (LIP) was injected intraperitoneally in rats in the TLE group, and saline instead of pilocarpine was treated in the control group. Six weeks later, rats were received 7.0-tesla T MRI scanning for structural and functional images. The volume of hippocampal gray matter volume and dorsal hippocampus related functional connectivity were compared between the two groups. Results Immediately after LIP treatment, 12 rats in the TLE group exhibited status epilepticus. Six weeks later, spontaneous seizures were observed in 10 of the 12 rats in the TLE group were observed spontaneous seizures. The volumes of hippocampal gray matter volumes in the TLE and control groups were 102.51 ± 1.97 mm3 and 85.76 ± 1.62 mm3, respectively; significant differences wasere noted between the two groups ( P 〈 0.05 ). As compared to controls, the rats in the TLE group revealed decreased dorsal hippocampus related connectivity to the ipsilateral amygdala and the ipsilateral ventral hippoeampus (P 〈 0.05 ), and increased connectivity to the contralateral cortex( P 〈 0.05 ). Conclusions In choronic LIP- TLE rats, hippocampal gray matter structures wereas impaired., and The functional connection in hippocampus-limbic system was diminished, while that in and hippocampus-cortex functional networks was enhancedere also abnormal.
