摘要 建立了一种基于CdTe量子点标记技术测定中性粒细胞明胶酶相关载脂蛋白(NGAL)的免疫层析法。用水相法制备的CdTe量子点作为荧光探针标记于NGAL的一株单抗,采用双抗体夹心法,制备了检测NGAL的层析试纸条,用于测定NGAL的含量。NGAL的线性范围为41-2050μg·L^-1,检出限为27.35μg·L^-1。对NGAL不同浓度的质控品连续测定10次,测定值的相对标准偏差在3.6%-9.7%之间。方法应用于尿液样品的分析,结果与酶联免疫吸附测定法对照,两种方法所得结果间无显著偏差。 Immunochromatographic test strips for the determination of neutrophil gelatinase-associated lipocalin (NGAL) was developed based on CdTe quantum dots as fluorescence probe. CdTe quantum dots prepared in aqueous medium were used as fluorescence probe and labelled on mono-antibody of neutrophil gelatinase-associated lipocalin, which were used in the preparation of fluorescence immuno-chromatographic test paper strips by the double-antibody sandwich technique for use in the determination of NGAL. Linearity range of NGAL was found between 41 μg·L^-1 and 2 050μg·L^-1 with detection limit of 27.35 μg·L^-1. Precision of the method was tested at different concentration levels of NGAL for 10 determinations, values of RSD's found were in the range of 3.6%-9.7%. The proposed method was applied to the analysis of urine samples, and checked by parallel analysis using the conventional method of enzyme-linked immunoadsorption, and no significant deviation was found between the results of the 2 methods.
