食管鳞癌中MMP-10和RKIP的表达与预后的关系 被引量:2

摘要 目的:分析基质金属蛋白酶10(MMP-10)和Raf激酶抑制蛋白(RKIP)在食管鳞癌中的表达,并探讨二者与临床病理特征及预后的关系。方法:采用免疫组化SP法检测92例食管鳞癌组织、相应癌旁组织以及10例正常食管组织中MMP-10和RKIP的表达,分析其表达与食管鳞癌临床病理参数的关系和二者相关性,并做出预后分析。结果:MMP-10在癌组织中的阳性率明显高于癌旁组织和正常食管组织,RKIP在癌组织中的阳性率明显低于癌旁组织和正常食管组织,MMP-10的表达与淋巴结转移以及TNM分期有关(P<0.05),而RKIP的表达与淋巴结转移有关(P<0.05);MMP-10和RKIP的表达呈负相关(r=-0.372,P=0.000);MMP-10阳性组的生存率低于阴性组(P<0.01),RKIP阴性的生存率低于阳性组(P<0.01)。Cox回归分析得出淋巴结转移、MMP-10阳性表达以及RKIP阴性表达均可作为食管鳞癌患者的独立预后因子。结论:MMP-10和RKIP与食管鳞癌的发生、发展、侵袭、转移以及预后相关,RKIP可能通过下调MMP-10的途径参与到食管鳞癌的侵袭、转移。 Objective To explore the expression of MMP-10 and RKIP in esophageal cancer,and to study their association with clinicopathologic features and prognosis. Methods the expression of MMP-10 and RKIP was detected in 92 esophageal squamous cancer tissues,their adjacent normal tissues and normal esophageal tissues by immunohistochemistry SP method, analyse the relationship of their expression with clinicopathologic features. Meanwhile, analyse their correlation and make prognosis analyse. Results the expression of MMP-10 was higher in esophageal squamous cancer than in adjacent normal tissues and normal esophageal tissues, which was positive correlated with TNM stage and lymph node metastasis. The expression of RKIP was lower in esophageal squamous cancer than in their adjacent normal tissues and normal esophageal tissues,which was negative correlated with lymph node metastasis. The expression of RKIP and The expression of MMP-10 were negatively correlated. The survival rates in positive expression group of MMP-10 and negative expression group of RKIP were significantly lower. In Cox model analysis showed that lymph node metastasis,the positive expression of MMP-10 and the negative expression of RKIP could acted as an independent prognostic factor. Conclusion MMP-10 and RKIP were related to tumorigenesis,progression,invasion and metastasis of esophageal squamous cancer. And RKIP may mainly down-express MMP-10 to participate the invasion and metastasis of esophageal squamous cancer.
