摘要 目的:探讨毛蕊花糖苷对阿尔茨海默病小鼠的神经保护作用。方法:将10月龄APP/PS1双转基因雄性小鼠随机分为模型组,熟地黄低、中、高剂量组(30、60、120 mg·kg-1),同龄C57BL/6小鼠为对照,每组8只。利用Morris法观察各组小鼠学习记忆能力;TUNEL法检测各组神经细胞凋亡情况;Nissl法检测小鼠脑组织神经元尼氏体表达;Western blot法检测各组β淀粉样蛋白表达情况。结果:毛蕊花糖苷中、高剂量组能够显著改善小鼠学习记忆能力;能够增加神经元数量,减少神经细胞凋亡及β淀粉样蛋白沉积。结论:毛蕊花糖苷能够改善阿尔茨海默病小鼠的学习记忆能力,其可能是通过促进神经元存活、减少凋亡并减少Aβ沉积实现的。 Objective:To investigate the protective effects of acteoside on the nerves in Alzheimer's mice model. Methods:10-month old APP/PS1 mice were randomly divided into model group,acteoside low,medium,high dose groups(30 mg·kg^-1,60 mg·kg^-1,120 mg·kg^-1)(n = 8),with agematched C57BL/6J mice of the same genetic background as control group. The ability of learning and memory were detected by Morris. Neural cell apoptosis was detected by TUNEL kit. Neurons and Nissl body quantity were detected by Nissl stain. And βamyloid protein expression was detected by Western Blot. Results:Acteoside medium,high dose groups could obviously improve the ability of learning and memory in mice,increased the number of neurons,and reduce neural cells apoptosis and βamyloid protein aggregation. Conclusion:Acteoside could improved the ability of learning and memory in mice,it could be achieved through increasing the number of neurons and Nissl body quantity,and reducing neural cells apoptosis and βamyloid protein aggregation.
