摘要
合成了基于脱单芳氨基反应的一氧化氮荧光探针(5-氨基荧光素)并对其进行了结构表征。研究发现合成的5-氨基荧光素作为荧光探针能选择性识别NO。探针的荧光强度与NOC18浓度(0-8.0×10^-5mol/L)呈良好的线性关系,经1H NM R和H RM S实验证实了探针识别NO为脱氨基的机理,并在小鼠单核巨噬细胞白血病细胞(Raw 264.7)活细胞中实现了对NO的成像。
A new model via reductive deamination of the aromatic primary amine as fluorescent probe for NO was synthesized and characterized. The results showed that the probe exhibited the selectivity to NO. The fluorescence intensity of probe showed a good linear relationship to the concentration of NOC18 (0 -8.0 × 10^-5 mol/L). The deamination mechanism of probe with NO was confirmed using 1H NMR and HRMS spectra. What more, this probe has been applied to image intracellular NO in living Raw 264.7 murine maerophages.
