妊娠期糖尿病孕妇中FoxM1基因的表达及其与胰岛素的关系 被引量:2

Expression of FoxM1 gene and its relationship with insulin in pregnant women with gestational diabetes mellitus

摘要 目的探讨妊娠期糖尿病孕妇中FoxM1基因表达变化以及与胰岛素的关系。方法选择东莞市石碣医院产科门诊行产检的GDM孕妇、正常孕妇各100例作为研究对象,分别为研究组和对照组,采用放射免疫法检测两组孕妇血清胰岛素水平,并采用逆转录聚合酶链反应(RT-PCR)检测两组孕妇血液中FoxM1基因表达。结果确诊GDM时,研究组孕妇的血清胰岛素表达水平明显高于对照组孕妇(t=12.22,P<0.05);分娩后6 h,两组孕妇的血清胰岛素表达水平均下降,研究组孕妇的血清胰岛素表达水平仍明显高于对照组孕妇(t=13.67,P<0.05)。确诊GDM时,研究组孕妇血液FoxM1 mRNA表达水平(2.38±0.45)明显高于对照组孕妇(1.25±0.37)(t=19.40,P<0.05);分娩后6 h,两组孕妇血液FoxM1 mRNA表达水平均下降,研究组孕妇的血液FoxM1 mRNA表达水平(1.39±0.27)仍明显高于对照组孕妇(0.74±0.16)(t=20.71,P<0.05)。研究组孕妇血清胰岛素表达水平与其血液FoxM1基因表达呈正相关(r=0.56,P<0.05)。结论 FoxM1基因参与了GDM发病机制,且其表达与GDM孕妇血清胰岛素表达水平呈正相关。 Objective To explore the changes of FoxM1 gene expression in pregnant women with gestational diabetes mellitus ( GDM) and its relationship with insulin. Methods One hundred pregnant women with GDM and 100 normal pregnant women with antenatal examination in Department of Obstetrics, Shijie Hospital of Dongguan City were selected as the study group and control group respectively. Radioimmunoassay was used to determine the serum levels of insulin of the two groups, and RT-PCR was employed for detecting the expression of FoxM1 gene. Results On the diagnosis of GDM, the expression level of serum insulin in the study group was significantly higher than that in the control group ( t = 12. 22, P〈0.05). Six hours after delivery, the expression levels of serum insulin of the two groups both decreased, but that of the study group was still significantly higher than that of the control group (t = 13.67, P〈0.05). On the diagnosis of GDM, the expression level of FoxM 1 mRNA was significantly higher in the study group than in the control group ( (2.38±0.45) vs. ( 1.25±0.37), t= 19.40, P〈0.05). Six hours after delivery, the expression levels of FoxM1 mRNA of the two groups both decreased, but it was still significantly higher in the study group than in the control group ( ( 1.39±0.27) vs. (0.74±0.16), t = 20.71, P〈0.05). The expression level of serum insulin in the study group was positively correlated with FoxM1 gene expression ( r= 0.56, P〈0.05). Conclusions FoxM1 gene is involved in the pathogenesis of GDM, and its expression is positively correlated with the expression of serum insulin in pregnant women with GDM.
