摘要
目的探讨不同剂量黄连水煎液对正常状态和脾虚状态大鼠胃肠平滑肌运动的影响,为临床用药提供动物实验依据。方法将80只清洁级SD健康雄性大鼠随机分为正常组,正常黄连高、中、低剂量组,脾虚模型组,脾虚黄连高、中、低剂量组,每组10只。脾虚各组采用利血平腹腔注射法建立脾虚证大鼠模型,共造模14 d,正常各组常规饲养。造模结束后,正常和脾虚黄连高、中、低剂量组分别灌胃5,2.5,1.25 g/kg黄连水煎液,正常组和脾虚模型组给予等体积生理盐水灌胃,均持续14 d。观察各组大鼠一般情况,计算大鼠胃排空率和小肠推进率;体外分离大鼠胃窦、回肠平滑肌细胞,计算胃窦、回肠平滑肌细胞收缩率。结果正常黄连各给药组大鼠从给药第2周开始出现嗜睡、少动,饮食量减少,整体精神状态不佳。脾虚各组大鼠于造模第2周出现典型的懒动、眯眼、嗜睡、食量下降等现象;脾虚黄连各给药组大鼠从给药第2周起虚弱、蜷缩、眯眼和弓背现象明显减轻,整体状态好于脾虚模型组。造模及灌胃期间,各组大鼠体质量比较差异均无统计学意义(P均>0.05)。正常黄连各给药组大鼠胃残留率均明显高于正常组(P均<0.05),正常黄连中剂量组小肠推进率明显低于正常组(P<0.05);脾虚模型组大鼠胃残留率和小肠推进率均明显高于正常组(P均<0.05);脾虚黄连高、中剂量组大鼠胃残留率和小肠推进率均明显低于脾虚模型组(P均<0.05),脾虚黄连小剂量组胃残留率和小肠推进率均有降低趋势,但与脾虚模型组比较差异均无统计学意义(P均>0.05)。脾虚黄连高、中剂量组胃窦平滑肌细胞收缩率均明显低于脾虚模型组(P均<0.05),脾虚黄连低剂量组回肠平滑肌细胞收缩率明显高于脾虚模型组(P<0.05)。结论一定剂量黄连水煎液能够抑制正常大鼠胃肠运动功能,改善脾虚状态大鼠的胃肠功能紊乱情况。
Objective It is to discuss the influence of Coptis decoction in various doses on gastrointestinal smooth muscle movement on normal and spleen deficient state rats, and to provide animal experiment basis for clinical medication. Methods Eighty clean SD male rats were randomly divided into normal control group, normal Coptis high, medium and low dose groups, model control group, model Coptis high, medium and low dose groups, each group had 10 rats. Spleen deficiency rat model was constructed by intraperitoneal injection of reserpine for a total of 14 days. After modeling, rats of Coptis groups were fed with different doses of Coptis decoction which was 5,2.5,1.25 g/kg by gavage, normal group and model group were fed with equal volume of normal saline for another 14 days. observation on the general conditions were kept,and gastric emptying and small intestinal propulsion changes were calculated; Separating rat gastric antrum and ileum smooth muscle cells in vitro and the contraction changes of cells were observed. Results Compared with normal control group, Coptis groups rats showed drows- iness, less dynamic, reduced diet and poor mental state as a whole. Compared with spleen dificiency model group, weakness, curling, squinting and camber of rats in Coptis groups were significantly reduced. Compared with normal control group,no sig- nificant difference was showed on the weight of rats in Coptis groups. Compared with spleen dificiency model group,no signifi- cant difference was showed on the weight of rats in Coptis groups. Compared with normal control group, the gastric residual rate of high-, medium-, and low-dose groups increased significantly( P 〈 0.01,P 〈 0. 001 ) ;and compared with normal control group, intestinal propulsive ratio of medium-dose groups decreased significantly(P 〈 0.05). Compared with spleen dificiency model group, the gastric residual rate of high-, medium-dose groups decreased significantly ( P 〈 0.05 ) ;intestinal propulsive ratio of high-, medium-dose group decreased significantly ( P 〈 0.05 ). Compared with model group, shrinking percentage of gastric antrum smooth muscle cells in high-, medium-dose increased significantly, and shrinking percentage of ileum smoothmuscle cell in low-dose group increased significantly(P 〈 0. 05). Conclusion Coptis decoction can inhibit normal rats gastro- intestinal function, improve the gastrointestinal dysfunction state of spleen deficient rats.
