摘要
目的探讨黄连苍薏四味方及其配伍对2型糖尿病大鼠血糖和血脂的影响及其作用机制。方法随机选取8只正常雄性SD大鼠作为正常组,选取40只雄性SD大鼠饲喂高脂饲料4周后腹腔注射链脲佐菌素(STZ)诱导2型糖尿病模型。将造模成功的大鼠随机分成黄连苍薏四味组、黄连+苍术组、黄连+生薏仁组、黄连+黄柏组、模型组,每组8只。各给药组均灌胃给药,给药体积均为1 mL/100 g,连续给药21 d;模型组和正常组给予等体积的蒸馏水灌胃。各组每隔7 d检测1次血糖、体质量;末次灌胃后称体质量,取血检测总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)和空腹胰岛素、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、脂联素水平,计算胰岛素敏感程度。结果黄连苍薏四味方组和黄连+苍术组血糖、TC、TG、LDL-C、TNF-α和IL-6水平均明显低于模型组(P均<0.05),空腹胰岛素和脂联素水平均明显高于模型组(P均<0.05);黄连+生薏仁组和黄连+黄柏组血糖、TC、TG、LDL-C水平均明显低于模型组(P均<0.05);黄连+黄柏组空腹胰岛素和脂联素水平均明显高于模型组(P均<0.05)。黄连苍薏四味方组血糖、IL-6、TNF-α水平均明显低于其他给药组(P均<0.05),空腹胰岛素水平明显高于其他给药组(P均<0.05),TC、TG、LDL-C水平明显低于黄连+生薏仁组和黄连+黄柏组(P均<0.05),脂联素水平明显高于黄连+生薏仁组和黄连+黄柏组(P均<0.05)。结论黄连苍薏四味方治疗糖尿病的机制与纠正脂代谢紊乱、抑制炎症因子表达、升高脂联素水平、改善胰岛抵抗、增加胰岛素分泌有关,且黄连苍薏四味全方治疗糖尿病的作用明显优于黄连苍薏四味方以黄连为君药的各组方。
Objective It is to investigate the effect of Huangliancangyifang on the blood glucose and blood lipids in type Ⅱ diabetic rats and its mechanism. Methods Eight male SD rats were selected as normal group, the other 40 male SD rats were selected to establish rat models of type Ⅱ diabetes mellitus by STZ and high fat diet, The successful rat models were randomly divided into Huangliancangyifang group, Berberine + Atractylodes group, Berberine + Coix seed group, Berberine + phelloden- dron group and model group, each group had 8 rats. All the rats in every medication group were treated at a dose of 1 mL/100 g through gavage for 21 days, and the ones in model group and normal group were given distilled water. The changes of blood glucose and weight of the rats were measured every 7 days. After the last gavage, body weight was measured, the rats'blood was gotten to determine the levels of TC, TG,LDL-C, IL-6, TNF-α, ADPN of the four groups, the insulin sensitivity was e- valuated. Results The levels of blood glucose, TC, TNF-α and TG, IL-6 in Huangliancangyifang group, Berberine + Atrac- tylodes group were significantly lower while that of fast insulin and ADPN were higher than that in model group( P 〈 0.05 ) ; the levels of blood glucose were significantly lower in Berberine + Coix seed group and Berberine + phellodendron group than that in model group ( P 〈 0, 05 ) , the level of fast blood glucose and ADPN in Berberine + phellodendron group was significantly higher than that in model group(P 〈 0.05). Conclusion The anti-diabetic mechanism of Huanglianeangyifang was related to regula- ting lipid metabolism,inhibiting expression of inflammatory factors, increasing the expression of ADPN, improving insulin re- sistance and insulin secretion. The anti-diabetic effect of Huangliancangyifang was significantly better than the prescription consisted of two herbs with the King' s drug of Berberine.
