摘要 目的采用HPLC变波长法同时测定金银花中绿原酸和芦丁的含量。方法利用高效液相色谱法测定,进样条件为:Zorbox SB-C18柱(4.6×150 mm,5μm),以乙腈-0.2%磷酸为流动相,梯度洗脱,流速1.0 mL/min,柱温为25℃,检测波长检测波长327 nm(0~20 min)、369 nm(20~32min),进样量为10μL。结果不同地区金银花中绿原酸和芦丁的含量差别较大,其中四川广汉地区所产金银花中绿原酸含量最高,河南地区所产金银花中芦丁含量最高,而山东地区所产金银花中绿原酸含量最低,川银花中芦丁含量最低。结论该实验方法简单方便,可以同时测定出金银花中绿原酸和芦丁的含量,实验条件稳定、可行,实验结果准确可靠,为金银花的质量控制提供了一定得参考。 Objective: simultaneous determination of chlorogenic acid and rutin. Methods: using the high performance liquid chromatographic method for the determination of chlorogenic acid and rutin. Chromatographic conditions: Zorbox SB-C18(4.6×150 mm, 5 um) column, using acetonitrile(pure)-0.2 % phosphoric acid analysis as mobile phase, flow rate is 1.0 mL/min. The detection wavelength was 327 nm(0~20 min), 369 nm(20~41 min), column temperature for 25 ℃, the sample volume was 10 u L. Results: the content of rutin and the content of chlorogenic acid in Lonicera japonica from different regions is different. The content of chlorogenic acid in sample of Sichuan is the highest, and the content of rutin in sample of Henan area was the highest, the content of chlorogenic acid in sample of Shandong area is the lowest, and the content of rutin in sample of Sichuan is the lowest. Conclusion: the method is simple and convenient, can also determine the content of chlorogenic acid and rutin, experimental conditions are stable, feasible, and the experimental results are accurate and reliable. This study can provide references for the quality control of Lonicera japonica.
