香蕉低聚糖的微波、超高压法提取与柱层析分离纯化 被引量:3

Extraction of Banana Oligosaccharides Using Microwave,Ultrahigh Pressure Methods and Purification by Column Chromatography

摘要 以香蕉为原料,研究微波法与超高压法提取香蕉低聚糖的工艺,并通过正交试验优化2种提取方法。比较Sephadex G-25葡聚糖凝胶柱层析和DEAE Sepharose Fast Flow琼脂糖凝胶柱层析对香蕉低聚糖提取液的分离纯化效果。结果显示:微波法提取低聚糖的优化工艺为:微波功率462 W,微波时间2.5 min,料液比1∶15 g/m L;超高压法提取香蕉低聚糖的优化工艺为:压力300 MPa,保压时间15 min,料液比1∶2 g/m L。微波法提取香蕉低聚糖的得率【17.72±0.49%】高于超高压法提取香蕉低聚糖的得率【8.05±0.03%】。Sephadex G-25葡聚糖凝胶分离香蕉低聚糖提取液的效果优于DEAE Sepharose Fast Flow琼脂糖凝胶,分离得到4个洗脱峰,表明香蕉中至少含有4种低聚糖。 The microwave and ultrahigh pressure methods on banana oligosaccharides extraction were studied. Sephadex G-25 and DEAE Sepharose Fast Flow column chromatography were used to separate and purify banana oligosaccharides extract. The processing of two methods were optimized by orthogonal experiments. Results showed that the optimum conditions of microwave method were as follows: microwave power 462 W, microwave time 2.5 rain, the ratio of material to solvent (W/V) was 1:15. And the optimum conditions of ultrahigh pressure were as follows: total ultrahigh pressure treating time was 2.5 min, ultrahigh pressure was 300 Mpa, the ratio of material to solvent (W/V) was 1:2. The yield of oligosaccharides extracted by the microwave method (17.72±O.49)% was higher than that of the ultrahigh pres- sure method (8.05±0.03)%. The seperation of Sephadex G-25 was better than DEAE Sepharose Fast Flow column chro- matography, and four elution peaks were isolated, which showed that bananas contain four kinds of oligosaccharides at least.
