摘要
目的研究高效氯氰菊酯(β-CP)对小鼠肝脏能量代谢的影响及黄芪多糖(APS)的保护作用。方法将50只雄性ICR小鼠随机分为5组,包括对照组(玉米油)、染毒组(β-CP 40 mg/kg)及不同剂量的保护组(在β-CP 40 mg/kg基础上分别加上100、200和300 mg/kg APS),连续灌胃染毒10 d。测定血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)的活力及肝脏组织匀浆中Na^+,K^+-ATPase、Ca^(2+),Mg^(2+)-ATPase和乳酸脱氢酶(LDH)的活力。结果与对照组比较,β-CP染毒组AST、ALT活力均显著升高,Na^+,K^+-ATPase、Ca^(2+),Mg^(2+)-ATPase、LDH活力均有所降低,差异均有统计学意义(P<0.05);与染毒组比较,各剂量保护组AST、ALT活力均有所降低,Na^+,K^+-ATPase、Ca^(2+),Mg^(2+)-ATPase、LDH活力均有所升高,其中,各剂量保护组AST、ALT、Na^+K^+-ATPase、Ca^(2+)Mg^(2+)-ATPase活力及200 mg/kg剂量APS保护组LDH活力与染毒组比较,差异均有统计学意义(P<0.05);各剂量保护组中200 mg/kg剂量保护组的Na^+,K^+-ATPase、Ca^(2+),Mg^(2+)-ATPase、LDH活力相对较高,更接近对照组水平。结论 40 mg/kg剂量β-CP对小鼠肝脏能量代谢有影响,APS对其具有保护作用,200 mg/kg剂量APS的保护作用较为明显。
[Objective]To explore the influence of beta cypermethrin(β-CP)on hepatic energy metabolism in mice and the protective effects of astragalus polysacharin(APS)on it.[Methods]Fifty ICR male mice were randomly divided into 5 groups,including the control group(corn oil),the experimental group(β-CP 40 mg/kg)and the protection groups(β-CP 40 mg/kg with APS100 mg/kg,200 mg/kg,300 mg/kg respectively). The mice were treated with the reagents through gavage daily for ten consecutive days. Afterwards,biomarkers including the AST,ALT in serum and Na^+,K~+-ATPase,Ca^2+,Mg^2+-ATPase,LDH in liver tissues were determined.[Results] Compared with control group,activities of AST,ALT were all increased while Na^+,K~+-ATPase,Ca^2+,Mg^2+-ATPase,LDH activities were all decreased in experimental group,all with significant differences(P〈0.05). Compared with the experimental group,activities of AST,ALT were all decreased while Na^+,K^+-ATPase,Ca^2+,Mg^2+-ATPase,LDH activities were all increased in protection groups. Among them,there were significant differences(P〈0.05)in Na^+,K^+-ATPase and Ca^2+,Mg^2+-ATPase activities in all 3 protection groups,and in LDH activity in the 200 mg/kg dose protection group. Comparison among 3 protection groups showed that activities of Na^+,K^+-ATPase,Ca^2+,Mg^2+-ATPase,LDH in the 200 mg/kg dose protection group were relatively high,closer to the level of the control group.[Conclusion]40 mg/kg doses of β-CP can influence hepatic energy metabolism in mice,and may be protected by APS,and the protective effect of 200 mg/kg dose of APS is more obvious.
