补肾调冲方对卵巢早衰大鼠性激素水平及TNF-α、IFN-γ蛋白表达的影响 被引量:21

Effect of Bushen Tiaochong Prescription on sex hormone level and TNF-α and IFN-γ protein expression in rats with premature ovarian failure

摘要 目的观察补肾调冲方对卵巢早衰模型大鼠血清促卵泡刺激素(FSH)、促黄体生成素(LH)、抗缪勒管激素(AMH)水平及卵巢组织肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)蛋白表达的影响。方法 60只SD雌性大鼠随机分为6组:对照组,模型组,补肾调冲方低、中、高剂量(以生药计24、12、6 g/kg)组,结合雌激素片(阳性药)组,每组10只。除对照组外,其余各组均ig雷公藤多苷片造模14 d,造模结束24 h后,给予相应药物治疗36 d。给药结束后,酶联免疫吸附(Elisa)法检测大鼠血清FSH、LH、AMH水平,免疫组化法检测卵巢组织TNF-α、IFN-γ蛋白表达。结果与对照组比较,模型组血清FSH、LH水平、卵巢TNF-α、IFN-γ蛋白表达均显著升高,AMH水平显著降低(P<0.05);与模型组比较,各给药组FSH、LH水平、卵巢TNF-α、IFN-γ蛋白表达均显著降低,AMH水平显著升高(P<0.01)。结论补肾调冲方可提高卵巢早衰大鼠卵巢储备能力,抑制卵泡封闭及黄体细胞凋亡,对卵巢早衰可能有治疗作用。 Objective To observe the effect ofBushen Tiaochong Prescription (BTP) on sex hormone level andTNF-α, IFN-γ protein expression in rats with premature ovarian failure. Methods Totally 60 SD female rats were randomly divided into six groups with 10 rats in each group: control group, model group, BTP low, medium, and high dose (24, 12 and 6 g/kg in crude drugs) groups, and Conjugated Estrogens Tablets (positive drug) group. Rats were ig administered with tripterygium glycosides for 14 d to make model of premature ovarian failure except for control group. Rats were treated with corresponding medicines for 36 d 24 h after that. Then the enzyme linked immunosorbent assay (Elisa) method was used to detect the levels ofAMH, LH, and FSH in serum, and the expression of TNF-α and IFN-γ in ovarian tissue was detected by immunohistochemistry. Results Compared with control group, the levels of serum FSH, LH and ovarian TNF-α ,IFN-γ protein expression of model group was increased significantly, and the level of serum AMH was significantly decreased (P 〈 0.05); Compared with model group, levels of FSH, LH, TNF-α and IFN-γ of BTP groups and Conjugated Estrogens Tablets group were significantly decreased, AMH level was significantly increased (P 〈 0.01) Conclusion BTP can improve the ovarian reserve, inhibit follicle blocking and apoptosis of corpus lutein cells of the premature ovarian rats, which suggests that BTP may have a therapeutic effect on premature ovarian disease.
