摘要
为准确、高效地利用分子标记辅助选择技术进行加工型辣椒胞质雄性不育(CMS)育性恢复基因的选择,本试验利用已报道的9个与辣椒恢复基因连锁的标记对加工型辣椒CMS不育系83-3A、恢复系812、F_1(83-3A×812)、F_1'(F_1×83-3B)与保持系83-3B回交群体进行试验,发现CRF-SCAR、PR-CAPS和OPP13-CAPS标记与CMS育性恢复基因Rf紧密连锁;用CRF-SCAR标记对BC_6F_1'的75个单株进行分子标记辅助选择验证,结果表明,39株有特异条带,与田间调查结果一致,正确率为100%;并从7份新引进未知育性材料中,检测到4份有恢复基因的材料。本试验结果表明,育种实际应用中可以利用标记CRF-SCAR对加工型辣椒恢复材料进行选择,能显著提高辣椒雄性不育三系育种的效率。
In ord er to accurately and effectively use molecular marker assisted selection technique in selecting restorer-of-fertility(Rf) gene in processing type pepper, 9 pairs of reported Rf gene linked markers were tested with the sterility line 83A-3A, the restorer line 812, F_1(83-3A×812), and the backcross population BC_6F_1' between F_1'(F_1×83-3B) and 83-3B in this study. The results indicated that CRF-SCAR, PR-CAPS and OPP13-CAPS markers were found closely linked to Rf gene. Screened by CRF-SCAR marker, in total of 75 individuals of BC_6F_1'population there were 39 individuals with the specific band, and the accuracy was 100% comparing with fertility observing in field. Using the CRF-SCAR marker 4 fertility restorers were selected from 7 newly introduced breeding materials. The results showed that CRF-SCAR markers can be applied to select restorer line of processing type pepper and it may significantly improve the efficiency of male sterility three-line breeding.
