摘要
本研究以转基因拟南芥和烟草叶片为材料,通过对DNA提取操作步骤及试剂用量进行了优化,探索出了一种简单高效的转基因拟南芥和烟草DNA提取方法。实验结果表明,本方法提取的拟南芥和烟草DNA条带清晰,完整,无降解,浓度较高,只有少量的蛋白质残留。利用PCR技术对提取的DNA样品进行检测,结果显示,所有的样品均能够扩增出目标条带,并且条带清晰。相比传统的拟南芥和烟草DNA提取方法,本方法具有简单,成本低,效率高,毒性低,时间短,提取DNA浓度高、纯度高等优点,适合于大批量转基因DNA的提取和鉴定。
The transgenic Arabidopsis thaliana and tobacco leaves were used as materials in this experiment. A simple and efficient method for DNA extraction was obtained by optimization the DNA extraction operation steps and reagent dosage. The results showed that the extracted DNA was clear in band, integral,no degradation,high concentration and with a small amount of protein residual. The extracted DNA as templates was used for PCR amplification to screen the transgenic lines, all the transgenic tobacco and Arabidopsis lines presented clear and exact bands of the target gene. Compared to conventional DNA extracted method of tobacco and Arabidopsis, the present method have many advantages,such as simple, low cost, high efficiency, low toxicity, short time,high concentration and purity. Therefore this method is suitable for large quantities of transgenic plants DNA extraction and identification.
