摘要
目的 为了更好地评价喹吡罗(QNP)诱导的大鼠强迫症模型的有效性和可靠性,探索强迫症的发生机制和治疗新方法。方法 健康雄性SD大鼠40只,随机分为四组,每组10只。QNP组,颈部皮下注射QNP(0.5mg/kg)每周2次;盐水组,每周2次,同体积的盐水代替QNP;QNP+CMI组,每周2次颈部皮下注射QNP,同时每天经腹腔注射氯丙咪嗪(CMI)10mg/kg;QNP+盐水组,采用等体积生理盐水代替CMI,其余处理相同。应用动物试验行为观测法和数字视频摄像量化分析法,研究试验动物的神经行为的改变。结果 大鼠在注射QNP5~7次后出现强迫性的出巢检查行为,CMI可以明显减轻这种行为。有的大鼠在注射QNP3~5次后会出现严重的反复啃噬行为。运动行为分析显示:大鼠在注射QNP10次后在开放的场地中返家的频率、运动的总距离明显高于对照组大鼠(P〈0.001),运动空间分布与对照组相比明显低于对照组(P〈0.001)。QNP组大鼠的运动距离和返家频率随注射QNP次数的增加而增加,但在第七次注射后,这些变化趋于平稳。结论 QNP可以诱导出大鼠的强迫检查行为,与人的强迫症行为类似,也有其他强迫行为如啃噬行为。神经行为量化分析更科学准确的验证了这一强迫症模型的有效性和可靠性。
Objective To evaluate the validity and liability of the animal model of obsessive - compulsive disorder (OCD) induced by quinpirole ,so we may propose a new approach for further understand the mechanism and therapy of OCD .Methods Forty naive male SD rats were randomly divided into four groups ,10 rats in each group .QNP group was treated by subcutaneous injection of Quinpirole hydrochlo‐ride (0 .5mg/kg) twice a week .The control group was treated with 0 .9% saline ,instead .QNP + CMI group was treated with the injection of Quinpirole and chlorimipramine (10mg/kg ) .QNP + saline group was treated with quinpirole and 0 .9% saline .Neural behavior of the rats was observed visually ,and vide‐oed by using the neural behavior observing system .The neural behavior data was quantitatively analyzed with the Ethovison progrom .Results After 5 ~ 7 times of QNP injection ,the rats had significant behavior of obsessive checking - home .This behavior induced by QNP could be relieved by chlorimipramine to some extent .Some rats performed behavior of obsessive biting after 3 - 5 times of QNP injection .The quantita‐tive tracking analysis showed that the rats treated by QNP moved much more distance in total ,and checked home more frequently .The moving distance and frequency of checking - home of QNP treated rats were in‐creased with the accumulation injection of QNP ,but kept at platue after seven times of injection .Conclu‐sions With QNP administration ,rats can be elicited to have obsessive checking - home behavior ,which is similar to humans OSD .Other behavior such as obsessive biting is a new finding in the present study and warrant further investigation .The quantitative analysis of neural behavior demonstrated the validity and li‐ability of the OCD animal model .
