摘要
目的:以RAW264.7泡沫细胞为研究对象,探讨黄芪多糖对胆固醇酯含量和ABCAl表达的影响。方法:将RAW264.7巨噬细胞诱导分化成泡沫细胞,用不同浓度黄芪多糖对其干预24h,酶法测定胆固醇酯含量,RT-PCR法检测泡沫细胞内ABCA1的mRNA表达。结果:对照组、10、20、50、100mg/L的黄芪多糖(APS)作用于泡沫细胞24h后,巨噬细胞内胆固醇酯含量分别为(45.15±3.57、37.67±2.26、34.59±3.51、28.57±10.34、15.92±3.86)nmol/mg。除10mg/L组外,其余各APS组与对照组比较,差异具有统计学意义(P<0.01)。采用RT-PCR方法检测ABCA1的mRNA表达,ABCA1与β-actin OD比值之比分别为0.41±0.03、0.59±0.03、0.64±0.08、0.77±0.11、0.95±0.07。根据其比值大小可判断各组ABCA1的相对表达水平。各APS组与对照组比较,差异具有统计学意义(P<0.01)。结论:黄芪多糖可能通过上调ABCAI的基因表达而降低泡沫细胞内胆固醇脂含量。
Objective:To investigate the cffect of astragalus poly-saccharides (APS)on cholesterol content and the expression of ABCA1 in RAW264.7 macrophage-derived foam cells.Methods:RAW264.7macrophages were incubated with oxidized low-density lipoproteins(ox-LDL)and rosiglitazone(0,10,20,50,100 mg/L),the cholesterol content of foam cells was measured by enzyme assay,ATP binding cassette transport A1(ABCA1)expression in the macrophages was quantitates by TR-PCR.Results:After APS of different concentrations were incubated with RAW264.7 foam cells for 24 h,the cholester-olester content of foam cells was (45.1 5±3.57,37.67 ±2.26,34.59 ±3.5 1,28.57 ±10.34,1 5.92 ±3.86)nmol/mg(P 〈0.01);ATP binding cassette transport A1(ABCA1)expression in the macrophages was quantitates by TR-PCR,OD ratio of ABCA1 andβ-actin were 0.41 ±0.03,0.59 ±0.03,0.64 ±0.08,0.77 ±0.1 1,0.95 ±0.07 (P 〈0.01 ).Conclusion:The increase in cholesterol efflux by APS might be relates to the up-regulation of ABCA1.
