摘要
目的优化手掌参中总皂苷的提取工艺并建立其含量测定方法,同时对其扰氧化活性进行研究。方法采用超声提取法,应用正交试验优化手掌参总皂苷的提取工艺,并以薯蓣皂苷元为对照,采用分光光度法测定总皂苷的含量。采用DPPH自由基清除法评价总皂苷的抗氧化活性。结果手掌参总皂忤最佳提取条件:95%乙醇提取、料-液比1:50(g/mL)、提取2次,每次90min,薯蓣皂苷元在6.14~21.49ug/mL浓度范同内与吸光度呈良好线性关系,平均回收率为103.6%,RSD=1.77%(n=6),总皂什含量存0.22%~1.09%。手掌参总皂苷对DPPH自由基有清除作用,且清除能力与总皂苷的含量有关。结论该提取工艺稳定,含量测定方法简便、快速、准确,手掌参总皂苷有一定的抗氧化活性。
Objective To optimize extraction technics of total saponins in Gymnadenia conopsea, to establish the method for determination of the content, and research its antioxidant activity. Methods The ultrasonic extraction method was used, the extraction process of total saponins was optimized by orthogonal design, and the content of total saponins was determinined by spectrophotometry with diosgenin as reference substance. The antioxidant, activities of total saponins was evaluated by DPPH free radical scavenging capacity. Results The optimum extraction conditions was 95% ethanol extraction, the ratio of material to solvent was 1:50 (g/mL), ultrasonic extraction for 2 times, every time was 90 minutes. The linear rang of diosgenin was 6.14-21.49 ug/mL, the average recovery was 103.6%, RSD=1.77%(n=6). The con- tents of total saponins were within 0.22%-1.09%. The total saponins had certain antioxidant activities and presented certain dose-effect relationship. Conclusion The extraction process is stable, the method for determination of content is simple, rapid and accuracy. The total saponins has certain antioxidant activities.
