Expression of microRNA -196a- 1 in gastric cancer and its clinical significance

摘要 目的探讨微小RNA(miRNA,miR)-196a-1在胃癌组织中的表达及其临床意义。方法采用反转录.聚合酶链反应(RT-PCR)检测miR-196a-1在胃癌组织和胃癌细胞株中的表达水平,并分析其与临床病理特征关系。结果RT-PCR结果显示,miR-196a-1在胃癌组织标本的表达量较对照组增加4倍,差异有统计学意义(P〈0.05);相对于正常胃黏膜上皮细胞株GES-1,胃癌细胞株SGC-7901、AGS、MKN28中的miR-196a-1均高表达,相对表达量分别为:SGC-7901:4.24±0.15(P〈0.05),MKN28:3.17±0.11(P〈0.05),AGS:3.32±0.07(P〈0.05)。胃癌组织miR-196a-1的高表达与患者的肿瘤TNM分期(P〈0.05)、淋巴结转移(P〈0.05)、组织分化程度(P〈0.05)有关,与患者的年龄、性别无关(P〉0.05)。结论miR-196a-1在胃癌组织和胃癌细胞中表达均上调,与胃癌的发生发展明显相关。 Objective To discuss the expression of microRNA ( miRNA, miR) - 196a - 1 in gas- tric cancer and its clinical significance. Methods The expression of miR - 196a - 1 in gastric cancer and gastric cancer cell lines was detected by reverse transcriptase -polymerase chain reaction (RT -PCR) and its relationship with clinicopathological features was analyzed. Results The expression of miR - 196a - 1 in gastric cancer tissue was increased by 4 - fold compared with that of the control group (P 〈 0. 05 ) , the difference was statistically significant (P 〈 0. 05 ); compared with normal gastric epithelial cell line GES - 1, human gastric cancer cell line SGC - 7901, AGS, MKN28 in miR - 196a - 1 were highly ex- pressed. Relative expression levels were: SGC - 7901 : 4. 24 ± 0. 15 ( P 〈 0. 05 ), MKN28 : 3. 17 ± 0. 11 ( P 〈 0. 05 ) , AGS : 3.32±0. 07 ( P 〈 0. 05 ). The expression of miR - 196a - 1 in gastric cancer tissue was related to TNM staging ( P 〈 0. 05 ), lymph node metastasis ( P 〈 0. 05 ) and the degree of histological differentiation ( P 〈 0. 05 ), which was not related to the age and sex of the patients ( P 〉 0. 05 ). Conclu- sion The expression of miR - 196a - 1 in gastric cancer and gastric cancer cells was up - regulated, which was significantly related to the occurrence and development of gastric cancer.
