白细胞介素-10基因多态性与肿瘤患者化学治疗后乙型肝炎病毒再激活的关系

Relationship between interleukin-10 gene polymorphism and hepatitis B virus reactivation in tumor patients after chemotherapy

目的探讨白细胞介素-10(IL-10)基因启动子区域-592A/C、-1 082G/A位点单核苷酸多态性(SNP)与肿瘤患者化学治疗后乙型肝炎病毒(HBV)再激活的关系。方法采用聚合酶链反应-限制性片断长度多态性(PCRRFLP)分析法,检测80例携带HBV的肿瘤患者的IL-10基因启动子-592A/C、-1 082G/A多态性,并确定其基因型和等位基因频率分布。结果 80例携带HBV的肿瘤患者,化学治疗期间出现HBV再激活18例(HBV激活组),62例患者未发生HBV再激活(HBV未激活组),HBV再激活发生率为22.5%。2组患者IL-10启动子区-592A/C基因型和等位基因频率比较差异均无统计学意义(P>0.05)。2组患者IL-10启动子区-1 082G/A位点SNP分析发现仅存在AA和AG基因型,而无GG基因型。HBV激活组患者IL-10启动子区-1 082G/A位点AA基因型及A等位基因频率显著高于HBV未激活组(P<0.05)。结论携带HBV的肿瘤患者化学治疗后HBV再激活与IL-10基因启动子区域-1 082G/A位点AA基因型及A等位基因有关。

Objective To investigate the relationship between the hepatitis B virus（HBV） reactivation and single nucleotide polymorphism（SNP） of interleukin-10（IL-10） gene promoter region -592A/C and -1 082G/A in tumor patients after chemotherapy. Methods The polymorphisms of the position -592A/C and -1 082G/A in IL-10 gene promoter region were determined by polymerase chain reaction-restriction fragment length polymorphisms（PCR-RFLP） analysis in 80 tumor patients, and the genotype and allele frequency distribution were determined. Results Among the 80 tumor patients with HBV, HBV was reactivated in 18 cases （HBV reactivation group）, and HBV wasn＇t reactivated in 62 cases （HBV nonactivated group）, the HBV reactivation rate was 22.5%. There was no significantly in the genotype and allele frequency of IL-10 gene promoter -592A/C between the two groups（P 〉0.05）. The SNP of IL-10 gene promoter region -1 082G/A showed that there was only AA and AG genotypes,hut no GG genotype. The genotype and allele frequency of IL-10 gene promoter region -1 082G/A in HBV reactivation group were signifieantly higher than those in HBV nonactivated group（ P 〈 0.05 ）. Conclusion HBV reacti- vation is associated with the -1 082G/A AA genotype and A allele in IL-10 promoter region in tumor patients with HBV after chemotherapy.
