摘要
目的研究确定建立长波紫外线(UVA)照射人表皮角质形成细胞(HEKC)损伤模型的照射剂量。方法选用原代培养的处于指数生长期的HEKC,将采用UVA剂量为3.07、6.14、12.29、24.58 J进行照射的设为剂量组,对未进行UVA照射的设为对照组。细胞损伤程度采用改良四甲基偶氮唑盐(MTT)法进行检测。结果 HEKC采用3.07 J UVA照射后,细胞损伤率7.27%;HEKC采用6.14 J UVA照射后,细胞损伤率32.73%;HEKC采用12.29 J UVA照射后,细胞损伤率72.73%;HEKC采用24.58 J UVA照射后,细胞损伤率95.45%;各剂量组吸光度(OD)值及损伤率与对照组比较差异有统计学意义(P<0.05)。结论建立UVA损伤HEKC模型最适照射剂量的IC_(50)值为8.09 J。
Objective To research and determine exposure dose in ultraviolet A (UVA)-damaged human epiderm keratinocytes (HEKC) model. Methods HEKC during exponential growth phase in primary culture were taken. Dosage groups included UVA exposure by doses of 3.07, 6.14, 12.29 and 24.58 J, and control group included non-exposed cases. Methyl thiazolyl tetrazolium (MTT) test was applied to detect cellular damage degree.Results After 3.07 J UVA exposure, HEKC had cellular damage rate as 7.27%; After 6.14 J UVA exposure, HEKC had cellular damage rate as 32.73%; After 12.29 J UVA exposure, HEKC had cellular damage rate as 72.73%; After 24.58 J UVA exposure, HEKC had cellular damage rate as 95.45%. The differences of optical delnsity (OD) value and damage rate between each dose group and control group had statistical significance (P〈0.05). Conclusion The optimal dose in establishment of UVA-damaged HEKC model is 8.09 J in ICs0.
