红松球果提取物对模拟失重诱导成骨细胞功能下降的防护作用 被引量:4

Protective Effects of Extracts of Pinus Koraiensis Cone on Dysfunction of Osteoblasts Induced by Simulated Weightlessness

摘要 目的从天然产物中寻找到对模拟失重诱导成骨细胞功能下降具有防护作用的提取物,为治疗失重性骨丢失提供潜在药物。方法从120多种天然产物中筛选出11种进行研究,筛选出对成骨细胞增殖活性具有较强效果的天然产物;在模拟失重下,将筛选出的天然产物作用于成骨细胞,并通过其对成骨细胞功能的影响评价其在防护骨丢失上的作用。结果红松球果提取物(extract of pine cone polyphenol,EPCP)显示出最强的成骨细胞增殖活性。模拟失重下,不同剂量的松树球果提取物对成骨细胞周期、细胞凋亡和碱性磷酸酶活性分别显示出了不同的防护作用。其中高剂量组可以显著提高模拟失重下成骨细胞碱性磷酸酶的活性,促进细胞周期运转并且抑制细胞凋亡。结论通过对天然产物促成骨细胞增殖活性进行筛选,所获得的EPCP对失重诱导的成骨细胞功能下降具有良好的防护作用。 Objective To develop potent bone loss protectant from natural products for weightless environment. Methods Eleven natural products selected from 120 plants were studied. The natural product with the highest proliferative activity in osteoblasts was picked out. Then the protective effect of the natural product on damages of osteoblasts induced by simulated weightlessness was evaluated and the impact of the natural product on oste- oblasts was studied. Results Extract of pine cone polyphenol (EPCP) showed the strongest proliferation activi- ty in osteoblasts. Under simulated weightlessness environment, different doses of EPCP showed different levels of protective effect on the cell cycle, apoptosis and activity of ALP in osteoblasts. In the high dose group, the activity of ALP was increased significantly, the cell cycle was promoted and the apoptosis was reduced. Con- clusion The extract of pine cone which was found to have high proliferative activity on osteoblasts, has protec- tive effect on damages of osteoblasts induced by simulated weightlessness.
