不同外植体和植物激素对大蒜愈伤组织诱导的影响 被引量:1

摘要 以大蒜鳞芽基部、茎尖、根尖和试管苗叶片为外植体,研究不同激素配比对大蒜愈伤组织诱导的影响。结果表明:大蒜根尖和鳞芽基部比茎尖和试管苗叶片易于诱导愈伤组织,根尖愈伤组织诱导率最高。不同激素配比对4种外植体愈伤组织诱导的影响不同,根尖在6-BA和2,4-D不同激素配比下全部诱导出愈伤组织;鳞芽基部愈伤组织诱导的最适激素组合为0.3 mg/L 6-BA+1.5 mg/L 2,4-D或0.5 mg/L6-BA+2.0 mg/L 2,4-D,诱导率为100%;茎尖愈伤组织诱导的最适激素配比为0.7 mg/L 6-BA+1.5mg/L 2,4-D,诱导率为85.83%;试管苗叶片愈伤组织诱导的最适激素配比为0.3 mg/L 6-BA+2.0 mg/L2,4-D,诱导率为71.67%。 The effects of different plant hormones on the callus induction of garlic were studied by using the base of clove,shoot tips,root tips and leaves of in vitro plantlet as explants in this study. The results showed that root tips and the base of clove were easier to induce callus than shoot tips and leaves of in vitro plantlet. And the callus induction rate of root tips was the highest in the four kinds of explants. Different plant hormone combinations had different effects on the four kinds of explants. The calli were induced from root tips in all combination of 6-BA and 2,4-D. The callus induction rate of the base of clove was 100% in the combination of 0. 3 mg / L 6-BA + 1. 5 mg / L 2,4-D,or 0. 5 mg / L 6-BA + 2. 0 mg / L 2,4-D. The callus induction rate of shoot tips was 85. 83% in the best combination of 0. 7 mg / L 6-BA and 1. 5 mg / L 2,4-D.The combination of 0. 3 mg / L 6-BA and 2. 0 mg / L 2,4-D was the best for leaves of in vitro plantlet to induce callus,and the rate of callus induction was 71. 67%.
