复方灵芝口服液对睡眠剥夺大鼠学习记忆的影响及机制 被引量:2

Effect and mechanism of compound Ganoderma lucidum oral liquid on learning-memory ability in sleep deprived rats

摘要 目的探讨复方灵芝口服液对睡眠剥夺大鼠学习记忆的影响及机制。方法雌性SD大鼠50只,随机分为5组(n=10):正常对照组(C组)、睡眠剥夺模型组(M组)、复方灵芝口服液低(0.15 g/kg,GL组)、中(0.75 g/kg,GM组)、高剂量(1.5 g/kg,GH组)干预组。腹腔注射氯苯丙氨酸(PCPA)制造睡眠剥夺模型,Morris水迷宫检测大鼠的学习记忆能力,比色法检测血清MDA含量及SOD活力,Western Blot法检测大鼠脑组织海马区p-p38MAPK蛋白的表达。结果根据行为学观察确定大鼠睡眠剥夺模型的成功。Morris水迷宫实验中,M组大鼠穿越平台的次数较正常对照组明显减少(P<0.05),而GH组寻找平台的潜伏期较M组明显缩短(P<0.05)。血清SOD活力和MDA含量的检测结果显示,与C组相比,M组大鼠血清SOD活力明显减少(P<0.05)、MDA含量明显增加(P<0.05);与M组相比,复方灵芝口服液各剂量组血清SOD活力均明显增加(P<0.05),而MDA含量均明显减少(P<0.05)。脑组织海马区p-p38MAPK蛋白表达检测结果表明,M组较C组明显增加(P<0.05),而复方灵芝口服液各剂量组p-p38MAPK蛋白的表达比M组均明显减少(P<0.05)。结论复方灵芝口服液可改善睡眠剥夺大鼠的学习记忆功能,其机制可能与减弱氧化应激和抑制P38MAPK信号通路有关。 Objective To observe the effects of Compound Ganoderma lucidum Oral Liquid( CGLOL) on learning-memory ability of sleep-deprived rats,and explore the underlying mechanism. Methods Female SD rats were randomly divided into five groups of 10 each,they were group C( control group),group M( sleep deprivation model group) and CGLOL intervention group [group GL( 0. 15 g/kg),group GM( 0. 75 g/kg),group GH( 1. 5 g/kg) ]. After the model was established by receiving parachlorophenylalanine( PCPA)( 300 mg/kg,i. p for 2 days.),Morris water maze was used to evaluate the learning-memory ability of rats,the content of serum MDA and the activity of serum SOD were determined by spectrophotometry,and the expression of p-p38 MAPK protein by Western Blot. Results According to the observation of ethological indexes,the sleep deprivation model was successfully established. In Morris water maze,group M crossed the platform less times than group C( P〈0. 05),group GH spent shorter time finding the platform than group M( P〈0. 05); According to the results of serum SOD activities and the content of serum MDA,the serum SOD activities decreased markedly in group M( P〈0. 05),while the content of serum MDA increased( P〈0. 05); When compared with group M,the serum SOD activities increased in CGLOL intervention group,while the content of serum MDA decreased( P〈0. 05). What's more,when compared with group C,the expression of p-p38 MAPK protein in rat hippocampus increased significantly in group M( P〈0. 05),but decreased in CGLOL intervention group when compared with group M( P〈0. 05). Conclusion Compound Ganoderma Lucidum Oral Liquid may ameliorate the effect of learning-memory ability caused by sleep deprivation; these effects may be mediated by removing free radicals and inhibiting P38 MAPK signaling pathway,and may be one of underlying mechanisms for Compound Ganoderma Lucidum Oral Liquid to relieve sleep deprivation
