蚓激酶对正常与软脂酸诱导的人肝细胞增殖的影响

摘要 目的:探讨不同浓度蚓激酶(EFE)在体外对LO2细胞株(人正常肝细胞)及软脂酸(PA)诱导的LO2细胞增殖的影响及EFE的最佳作用浓度。方法:用不同浓度(100 u/m L、200 u/m L、400 u/m L、600 u/m L、800 u/m L、1 000 u/m L)EFE作用于LO2细胞,分别在作用6 h、12 h、24 h,通过MTT法检测各组细胞的OD值;用20μg/m L的PA作用LO2细胞24 h,然后用不同浓度(100 u/m L、200 u/m L、400 u/m L、600 u/m L、800 u/m L、1 000 u/m L)的EFE进行干预,并于6 h、12 h、24 h检测细胞增殖情况。结果:200 u/m L、400 u/m L的EFE可促进LO2细胞的增殖,但无统计学意义,高浓度(800 u/m L、1 000 u/m L)的EFE能明显抑制LO2细胞的增殖(P<0.05),且其抑制作用随着时间的延长而增强;低浓度(100 u/m L)的EFE可促进经PA诱导的LO2细胞的增殖,其作用效果显著(P<0.05);200 u/m L、400U/m L的EFE对经PA诱导的LO2细胞的促进作用极显著(P<0.01)。结论:EFE能缓解PA诱导的细胞损伤,对PA诱导的LO2细胞具有保护作用,且其最佳作用浓度为400 u/m L。 To investigate effects of different concentrations of earthworm fibrinolytic enzyme(EFE) on proliferation of normal human liver cells LO2 cells in vitro and palmitic acid(PA) induced LO2 cells, and find out the optimal concentration of EFE, LO2 cells were treated with different concentration of EFE(100, 200, 400, 600, 800 and 1 000 u/m L), then MTT method was used to to determine the OD value after 6 h, 12 h and 24 h in different groups. Meanwhile LO2 cells induced by 20 μg/m L PA for 24 h were also treated with different concentration of EFE(100, 200, 400, 600, 800 and 1 000 u/m L) and the proliferation of LO2 cells were detected by MTT after 6 h, 12 h and 24 h. The results showed that EFE at the concentrations of 800 u/m L and 1 000 u/m L could significantly inhibit the proliferation of LO2 cells(P〈0.05), and the longer the time was, the stronger the inhibition was. As well,EFE at low concentration of 100 u/m L could significantly(P〈0.05) promote the proliferation of LO2 cells induced by PA, and EFE at the concentration of 200 and 400 u/m L could extremely significantly(P〈0.01) promote the proliferation of LO2 cells induced by PA. In conclusion, EFE can relieve damage of LO2 cells induced by PA, and the optimal concentration is 400 u/m L.
